大白猪Leptin基因外显子3多态性及其与部分经济性状的关系

摘　　要：本实验以大白猪为研究对象,利用PCR-RFLP法检测Leptin基因外显子3的多态性,进一步探讨其与生长、繁殖性能的相关性。结果表明:大白猪实验群体Leptin基因第3外显子存在T3469C突变,检测到CC、CT和TT 3种基因型,T为优势等位基因,且该位点处于Hardy-Weinberg平衡状态(P>0.05);Leptin基因外显子3 T3469C变异位点对繁殖性能具有显著影响,CC基因型个体的经产总产仔数显著高于CT和TT型个体(P<0.05),并且CC型个体的经产产活仔数也显著高于CT型个体(P<0.05);CC基因型个体的平均初生重和断奶重高于CT及TT型个体,且CC型个体100 kg体重日龄平均值低于CT和TT基因型个体,但差异不显著。研究结果显示,大白猪Leptin基因T3469C位点多态性对生长、繁殖性能均有不同程度的影响,CC基因型个体不仅具有较高的产仔性能,而且具备较快的生长速度。This experiment was conducted to analyze the genetic variations at exon 3 of Leptin gene by PCR-RFLP,further discussed the correlation between the different genotypes of Leptin gene and the growth,reproductive performance in Yorkshire pigs.The results showed that exon 3 of Leptin gene digested by Hinf Ⅰ could be divided into three kinds of genotypes （CC,CT and TT） in Yorkshire test groups,and the allele T was dominant one.This locus was in Hardy-Weinberg equilibrium （P〉0.05） in above population.Genetic variation of exon 3 mutations of Leptin gene had a remarkable impact on reproductive performance,correlation analysis showed that the individuals with CC genotype were significantly higher than those individuals with CT and TT genotypes in total number born of multiparity （P〈0.05）,and individuals with CC genotype were also significantly higher than those individuals with CT genotype in number born alive of multiparity （P〈0.05）.For production trait,individuals with CC genotype were higher than those individuals with CT,TT genotypes in the average value of birth weight and weaning weight,in addition,the average age at 100 kg of individuals with CC genotype were lower than the other two genotypes individuals ＇,while there were no significant differences among them.These results indicated that the polymorphisms of Leptin gene at T3469C had an effect on both the growth and reproduction performance of Yorkshire pigs in a way,the individuals with CC genotype not only had higher litter performance,but also had faster growth rate.
