摘 要:本实验研究旨在褪黑素对缓解热应激引起的雄性小鼠精液品质下降,以及受精后早期胚胎发育阻滞的作用.将8~10周龄健康雄鼠随机分为对照组(C)、对照加褪黑素组(C+M)、热应激组(H)、热应激加褪黑素组(H+M)4个处理.热应激组采用人工气候箱模拟夏季高温天气;对照组采用人工气候箱模拟适宜温度.热应激11d后对雄鼠体重、睾丸重、精子密度、顶体完整性、质膜完整性进行计数.在热应激1id及热应激17d后和雌鼠合笼,收集合子进行体外培养并观察其发育情况.结果表明:注射褪黑素的热应激公鼠相对于对照组的热应激公鼠,其睾丸指数(0.34vs.0.33)、精子的密度(3.5vs.3.3)、顶体完整性(72.2 vs.70.3)没有显著改善(P>0.05),精子膜完整性有所改善(45.71% vs.42.63%),体外培养胚胎卵裂率(89.34% vs.66.2%)和囊胚率(38.95%vs.2.32%)显著提高(P<0.05),囊胚细胞总数(49.4枚vs.38.6枚)显著增加(P<0.05),囊胚凋亡细胞数(2.6枚vs.4.5枚)显著减少(P<0.05),对公鼠注射褪黑素显著改善了后续胚胎发育的状况.In this study,we analyze whether the administration of melatonin to male mice would alleviate the adverse effect on semen quality and the subsequent early embryo development induced by heat stress.8-10 weeks healthy male mice were randomed selected into four groups:the control group(C),the conrol group with melatonin(C+M),the heated group (H) and the heated group with melatonin (H+M).The heated group and the heated group with melatonin were put into the the artificial climate chamber which imitate the summer temperature.The control group and the control group with melatonin were also put into an artificial climate chamber with appropriate temperature.After 11 days from the male mouse were put into the chamber,we examined the weight of the body and the testis,the density of the sperm,the intactness of the acrosome and the membrane of the sperm.On 11 days and 17 days from the male mouse were put into the chamber,female mouse were mated with male mouse and were cheked pregnant and then the female mouse with virginal plug were anaesthetized and the zygotes were collected to be cultured in vitro.The results of our experiment indicated that,the testis index (0.34vs.0.33),the densty of the sperm (3.5vs.3.3) and the intactness of the sperm acrosome (72.2vs.70.3;P〉0.05) derived from the heated group with melatonine were not significantly higher compared with the heated group,but the rates of cleavage (89.34%vs.66.2%; P〈0.05) and blastocyst (38.95%vs.2.32%; P〈0.05) of embryos derived from the heated group with melatonine were significantly higher compared with the heated group.The blastocyst from derived from the heated group with melatonine had more cells (49.4 vs.38.6; P〈0.05) and less apoptosis cells (2.6 vs.4.5; P〈0.05) compared with the heated group.These results suggest that administration of melatonin to heat stressed male mice has a better performance of the subsequent embryos development.
