摘　　要：研究中药复方提取物对小鼠乳腺上皮细胞增殖的影响,旨在为探明中药复方促进母畜泌乳量的机制提供试验依据。采用混合酶消化法分离成年妊娠中期的ICR小鼠乳腺上皮细胞,通过差酶消化法和相差贴壁法获得纯化后的乳腺上皮细胞,运用SDS-PAGE对纯化后细胞进行分析。使用水煎和醇提2种方法制备中药复方提取液,分别作用于对数生长期的乳腺上皮细胞1、3、5 d,使用MTT法检测细胞活力。结果表明:使用混合酶消化法与差酶消化法和相差贴壁法相结合可得到大量具有酪蛋白分泌功能的乳腺上皮细胞;中药复方提取物在高浓度时对乳腺上皮细胞的增殖有促进作用,在低浓度时对乳腺上皮细胞的增殖有抑制作用;促进作用随时间的增加而降低,抑制作用随时间的增加而增强。醇提的中药复方促进细胞增殖的浓度范围大于水煎的中药复方,而且醇提的中药复方作用效果远好于水煎的中药复方,可能是因为促进乳腺上皮细胞增殖的药物成份为醇溶性。The experiment was carried out to study the effects of Chinese medicinal herb extracts on proliferation of mice mammary gland epithelial cells.Mammary gland epithelial cells were separated from ICR mice in second trimester of pregnancy by using mixed enzyme digestion,epithelial cells were purified according to enzyme alternative digestion and differential adhesion method.Then the purified cells were analyzed by SDS-PAGE.Chinese medicinal herbs were extracted by water and alcohol and the extracts were added to cell cultural medium in logarithmic phase for one day,three days and five days,respectively.Then the viability of mice mammary gland epithelial cells was determined by MTT.The normal mice mammary gland epithelial cells which had function of casein excretion could be obtained by enzyme alternative digestion and differential adhesion method.Chinese medicinal herb extracts could promote the multiplication of mice mammary epithelial cells at a high concentration,but as the concentration was at a low level,the inhibiting effect appeared.The promoting function was weaker as the time increasing; in contrast,the inhibiting effect was stronger as the time increasing.Chinese medicinal herb extracts by alcohol had a larger concentration range than by water for promoting the proliferation of cells,and the effect of Chinese medicinal herbs extracted by alcohol was better than that extracted by water,maybe the component of drugs to promote the multiplication of the mice mammary gland epithelial cells are alcohol solubility.
