摘  要:目的:研究富硒板党(BCPA)注射液对老龄大鼠免疫功能的影响及其机制。方法:2月龄、12月龄和22月龄大鼠(雌雄各半)60只,分别作为年轻组、中年组、老龄组。每个年龄段大鼠随机分为对照组和BCPA实验组(n=10)。BCPA实验组每天腹腔按7.2 g/kg注射BCPA,对照组注射等量生理盐水,常规饲养连续45 d。BCPA给药后第15天,进行免疫注射。第22天,迟发型免疫应答(DTH)诱导。第15天、30天及45天尾静脉采集各年龄组大鼠血液,抗原特异性IgG、IgG1及IgG2α检测。第45天,取脾细胞分析主要T细胞亚群,并增殖脾细胞。结果:年轻、中年组BCPA实验组与对照组比较DTH反应均无显著差异,老龄BCPA实验组DTH反应则明显增强。特异性IgG、IgG1及IgG2α抗体年轻BCPA实验组与对照组比较无显著性差异,老龄BCPA实验组则明显增高(P<0.05)。CD3+T淋巴细胞、CD4+辅助T淋巴细胞在年轻组中比较无显著差异,中年、老龄组中有显著提升(P<0.05)。年轻BCPA实验组脾细胞有很强的增殖反应(P<0.05)。结论:BCPA能增强老龄大鼠DTH反应,增强抗KLH特异性IgG、IgG1和IgG2α抗体的产生,可提高对抗原的反应性,提高CD4+T细胞数量,促进免疫应答,在对抗免疫衰老,提高抗氧化能力方面有重要的作用。Objective: To study the effects of the Rich Selenium-Banqiao-Codonopsis Pilosula (BCPA) injecta on the aged rats' immune functions and its underlying mechanism. Methods: Totally 60 rats, composed of 2, 12 and 22 month age old (half male and half female), were served as a young group, middle- age group and aged group respectively. Each group rats were randomly divided into the control and the BCPA subgroup( n = 10). The BCPA group was injected with BCPA at 7.2 g/kg intraperitoneally every day and the control group was injected the same volume of normal saline. All rats were conventionally fed for 45 days. An immune injection was performed after 15 days of BCPA in- jection. On the 22nd day, late-onset immune response would be induced. The caudal vein blood was collected and the antigen specific IgG, IgG1 and IgG2α antibody was detected on the 15th, 30th and 45th day. On the 45th day, the major T cell subgroups of splenic cells were ana- lyzed and splenic cells were proliferated. Results: No significant difference in the delayed-type hypersensivity(DTH)reaction was found be- tween the control and the BCPA subgroups in the young and middle- aged rats while the aged BCPA subgroup had a stronger DTH reaction. There was no significant difference in the blood content of specific IgG, IgG1 and IgG2a antibody between the young and middle-age BCPA group'while the aged BCPA group rots had an obvious enhancing reaction to the three antibodies mentioned above ( P 〈 0.05). There was no obvious difference in the number of the CD3 + lymphocytes and the CD4 + T helper lymphecytes between the control and the BCPA subgroup in the young aged rots while a significant increase was spotted between the middle-aged and the aged group ( P 〈 0.05). The splenic cells from young BCPA group rots had a strong proliferation response ( P 〈 0.05).
