摘  要:目的探讨部分肝切除术对小鼠近期与远期学习记忆功能及海马炎性细胞因子表达的影响。方法将32只C57BL/6小鼠随机均分为早期手术组(O5组)、早期对照组(C5组)、远期手术组(O35组)和远期对照组(C35组)。对手术组进行部分肝切除术后,C5、O5组在术后第1-4天行Morris水迷宫测试,第5天采用RQ-PCR检测海马组织TNF-α和IL-6mRNA表达;O35、C35组在术后第31-34天行Morris水迷宫测试,第35天检测海马组织TNF-α和IL-6mRNA表达。结果与C5组比较,O5组的水迷宫测试成绩下降(P<0.05)、海马TNF-α和IL-6 mRNA表达增加(P<0.05)。O35组水迷宫测试成绩较O5组改善(P<0.05),但仍差于C35组(P<0.05);O35组的TNF-α和IL-6mRNA表达与O5组无统计学差异(P>0.05),但高于C35组(P<0.05)。结论部分肝切除术使小鼠术后近期和远期学习记忆功能下降,海马炎性细胞因子表达上调。Objective To investigate the effects of partial hepatectomy on short-term and long- term learning and memory function and the expressions of inflammatory cytokines in the hippocampus of mice. Methods Thirty-two male C57BL/6 mice were equally and randomly divided into four groups of O5 (early-term operation), C5 (early-term control), O35 (long-term operation) and C35 (long-term control). Morris water maze(MWM) was performed from the 1st day to the 4th day in groups of O5 and C5 or from the 31st day to the 34th day in groups of 035 and C35. The expressions of TNF-α and IL-6 mRNA in the hippocampus were determined by RQ-PCR on the 5th day in groups of O5 and C5 or on the 35th day in groups of O35 and C35 after partial hepatectomy. Results Compared with group C5 MWM score was decreased and the expressions of TNF-α and IL-6 mRNA in the hippocampus were increased in group O5 (P〈0. 05 ). MWM score was better in group O35 than that in group Os (P〈0. 05) ,which was worse in group O35 than that in group C35 (P〈0.05). There were no significant differences in the expressions of TNF-α and IL-6 mRNA in the hippocampus between group 035 and group O5 ( P〉0. 05 ), which were higher than those in group C35 ( P〈0. 05 ). Conclusion Partial hepatectomy may reduce the ability of learning and memory function with increased expressions of inflammatory cytokines in the hippocampus in mice.
