摘  要:目的 观察活血散瘀方对颅脑损伤大鼠神经功能缺损评分、脑组织含水量和水通道蛋白(AQP)4表达的影响。方法 SPF级SD大鼠100只按随机数字表法分为模型组和活血散瘀方组,每组各50只,均参照Feeney自由落体撞击法制作颅脑损伤模型。活血散瘀方组大鼠造模后应用消肿散瘀混悬液20mL/kg灌胃直至处死,模型组大鼠灌胃等剂量的蒸馏水。于造模后12h、ld、3d、5d、7d每组取10只大鼠进行神经功能缺损评分和脑组织含水量的测定,免疫组化染色检测脑组织AQP4的表达。结果 活血散瘀方组大鼠造模后12h、1d、3d、5d、7d神经功能缺损评分、脑含水量、AQP4的表达均低于模型组[3d:2.38±0.13vs2.98±0.24、(82.84±0.29)%vs(90.14±0.32)%、0.22±0.02vs0.25±0.03],差异有统计学意义(P〈0.05)。结论 活血散瘀方能改善创伤性脑损伤大鼠的神经功能缺损评分,可能与降低AQP4表达,抑制细胞内水肿有关。Objective To observe the effects of Xiaozhong Sanyu (XZSY) pills on neurologic severity scale scores, water content and aquaporin 4 (AQP4) expression in the brain of rats after traumatic brain injury (TBI). Methods One hundred SPF-grade SD rats were randomly divided into TBI model group and XZSY intervention group (n=50); TBI models were created by Feeney free-fall impact method. XZSY pills at 20 mL/kg by gavage were given to the XZSY intervention group, and equivalent dose of distilled water was given to the TBI model group; 12 h, and 1, 3, 5 and 7 d after TBI, neurologic severity scale was performed, water content in the brain was measured, and immunohistochemistry was used to analyze the AQP4 expression. Results As compared with those in the TBI model group, the neurologic severity scale scores, water content and AQP4 expression in the XZSY intervention injury were significantly decreased at all observation time points (3 d: 2.38±0.13 vs. 2.98±0.24, 82.84±0.29 vs. 90.14±0.32, 0.22±0.02 vs. 0.25±0.03, P〈0.05). Conclusion XZSY can protect the brain tissues after TBI by reducing AQP4 protein expression and inhibiting intracellular edema.
