摘  要:通过去壁低渗法染色体制片,探针标记制备、染色体DAPI浓度的优化,建立了枣基因组原位杂交(Genomic in situ hybridization,GISH)技术体系,并用该技术检测了枣和酸枣基因组之间的同源性。用阜平大枣做探针给酸枣杂交,用酸枣做探针给阜平大枣杂交,以及分别用阜平大枣、酸枣做探针给赞皇大枣杂交,所有染色体都显示出较强的杂交信号并均匀布满其全长。结果表明:枣和酸枣基因组之间具有高度同源性,为证实枣起源于酸枣,两者同属一个种提供了分子细胞遗传学的证据,研究也表明赞皇大枣为同源三倍体。Through optimization of the wall degradation hypotomic method of chromosome,probe preparation and chromosome DAPI concentration,agenomic in situ hybridization(GISH)technique was established in Ziziphus Mill.,and the genomic homology between jujube and wild jujube was checked by this technique.GISH of the wild jujube with genomic DNA probe of Chinese jujube‘Fupingdazao'on metaphase and GISH of‘Fupingdazao'with genomic DNA probe of wild jujube on metaphase,and GISH of Chinese jujube‘Zanhuangdazao'with genomic DNA probe of‘Fupingdazao'or wild jujube on metaphase showed very strong hybridization signals in all chromosomes and the signals distributed on the whole chromosomes.The results showed that Chinese jujube and wild jujube had high genomic homology,the new evidence was provided to confirm the jujube originated from wild jujube and they belonged to the same species.‘Zanhuangdazao'should be an autotriploid.
