摘 要:为了对瓜果腐霉PAM1菌株中的除草活性物质进行分离纯化和结构分析,本试验通过有机溶剂超声波提取、硅胶柱层析和高效液相色谱相结合的方法对除草活性化合物进行分离,利用质谱、红外光谱和核磁共振等技术对所得到的化合物进行了结构分析。结果表明:从瓜果腐霉PAM1菌株的乙酸乙酯粗提物、甲醇粗提物中分别获得了保留时间为16.37和8.55min的化合物1和化合物2,2个化合物对马唐叶片均表现出较强的除草活性。化合物1的核磁图谱显示其分子中含有不饱和键、羟基和羧基,质谱图提示其分子量为578,推断其可能的分子式为C37H70O4;化合物2可能的分子量为453。瓜果腐霉菌丝提取得到的2个化合物均为长链脂肪酸类化合物。The objective of this study is to separate and purify the herbicidal fractions produced by the mycelia of PAM1,an isolate of the pathogen Pythium aphanidermatum.The mycelia were extracted sequentially with ethyl acetate and methanol and then treated with ultrasonic,and the crude extracts were separated by using the methods of column chromatography and HPLC.The chemical structures of the compounds obtained were analyzed by using the techniques of mass spectra(MS),infrared spectrum(IR)and nuclear magnetic resonance(NMR).Compound 1and compound 2were obtained from the ethyl acetate and methanol extracts,respectively,which showed high bioactivity on Digitaria sanguinalis.The NMR spectrum of compound 1indicated that there were unsaturated bonds,hydroxyl group and carboxyl in its molecular,and the MS spectrum showed its molecular weight was 578,which in-dicated that the formula was C37H70O4.And the probable formula of compound 2was likely to be 453.The two compounds separated from the extracts of the mycelia of Pythium aphanidermatum were deduced to be a kind of fatty acids.
