摘  要:首先采用单因素实验法,寻找提取银杏叶粗多糖各影响因素的取值范围,进而采用均匀设计法优化银杏叶粗多糖的水提醇沉工艺,得到最佳提取工艺条件为:提取时间为210min,液固比为30∶1,提取温度为80℃,提取次数为2次,结果表明:在此条件下粗多糖得率最高为7.14%,与理论值6.96%非常接近。然后采用苯酚硫酸法测定每次试验的纯多糖含量,以纯多糖得率为指标对提取工艺进行进一步优化,得到银杏叶纯多糖的最佳提取条件为:提取时间为210min,液固比为1∶35,提取温度100℃,验证实验表明,试验得到的真实得率与理论值亦非常接近。由此可知,均匀设计试验优化银杏叶多糖提取条件,所得工艺参数是可靠的。Firstly,single factor experiment method was used to find the range of each influencing factor.Then,the extraction process of polysaccharides from Ginkgo biloba Leaves was optimized with Uniform Design.The best extraction process for crude polysaccharides was listed as follows:extraction time was 210 min,liquid-solid ratio was 30∶1(mL/g),extraction temperature was 80℃,and extraction times was twice.Under the optimal extraction condition,the highest extraction rate obtained was 7.14%.Verification results was very close to the theoretical value 6.96%.And then the phenol-sulfuric acid method was used to determine the polysaccharide content of each experiment and polysaccharide yield was chosen as indicators for further optimization of the extraction process too.Finally the best extraction conditions for pure polysaccharide was listed as follows:liqiud-solid ratio 35∶1,extraction time210 min,temperature 100℃,the verification results shows that the real yield and the theoretical values were also very close.It proved the process parameters obtained by Uniform Design was reliable.
