摘　　要：目的探讨野菊花总黄酮联合顺铂对人骨肉瘤MG-63细胞抑制作用及可能的机制。方法采用不同质量浓度野菊花总黄酮与顺铂单独及联合作用MG-63细胞;倒置相差显微镜观察细胞形态变化;CCK-8法检测细胞生长抑制率,并计算金氏公式Q值进行协同性分析;流式细胞仪和RT-PCR检测凋亡率及凋亡相关基因bcl-2、caspase-3和p21的表达情况。结果野菊花总黄酮与顺铂单独及联合作用均能够抑制MG-63细胞增殖,诱导其凋亡,并下调bcl-2及上调caspase-3、p21基因的表达;Q值提示两者低质量浓度联合作用具有协同效应,而较高质量浓度联合作用则表现为叠加效应,未显示出拮抗效应。结论野菊花总黄酮在诱导MG-63细胞凋亡起重要作用,但野菊花总黄酮中的单体发挥抗肿瘤作用和协同抗肿瘤的效果有待进一步深入研究。AIM To investigate the effect of total flavones from Chrysanthemum indicum combined with Cispla- tin on human osteosarcoma cell line MG-63 and its mechanism. METHODS MG-63 cells were treated with total flavones from Chrysanthemum indicum combined with or without Cisplatin. The cellular morphologic changes were observed under inverted phase contrast microscope. The effects of proliferation inhibition were assayed by CCK-8. The drug-drug interactions were judged through Q value calculated by King~ formula. The apoptosis ratios of cells were analyzed by flow cytometry. The expression of bcl-2, caspase-3 and p21 gene was detected by RT-PCR. RE- SULTS With or without Cisplatin, total flavones from Chrysanthemum indicum could inhibit the proliferation and induce the apoptosis of MG-63 cells while the expression of bcl-2 reduced and the expression of caspase-3 and p21 increased. The combination of total flavones from Chrysanthemum indicum and Cisplatin exerted a synergistic effect at low concentrations while a superimposed effect at high concentrations. CONCLUSION Total flavones from Chrysanthemum indicum play a key role in inducing the apoptosis of MG-63 cells. These results warrant further studies to clarify which constituent or constituents combination have a potential for anticancer effect.
