摘 要:目的研究锁阳多糖对过氧化氢(H2O2)诱导非洲绿猴肾上皮(Verda Reno,简称VERO)细胞损伤的保护作用。方法建立H2O2致VERO细胞氧化损伤模型,MTT法检测细胞存活率;通过荧光光度法检测caspase-3活性;利用荧光探针DCFH-DA对荧光染色的胞内活性氧(ROS)检测其荧光强度;化学比色法测定细胞中超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性,以及细胞培养液中丙二醛(MDA)水平。结果锁阳多糖能明显改善H2O2诱导的VERO细胞的氧化损伤,与模型组相比,锁阳多糖组可使细胞存活率升高,细胞内的ROS的积累量显著降低(P<0.01),caspase-3活性下降(P<0.05),同时细胞内的SOD和GSH-Px活性明显增加(P<0.01),细胞培养液中的MDA水平明显降低(P<0.01)。结论锁阳多糖能对H2O2诱导的VERO细胞的氧化损伤保护提示其具有潜在的防治肾脏相关疾病的作用。AIM To study the protective effect of polysaccharide from Cynomorium songaricum Ruhr on the oxidative damage of VERO ceils induced by H2O2. METHOD Cell viability was assayed by MTT method. The toxicity model was established by treating VERO cells with H2 O2. The caspase-3 activity was measured by chemical colorimetric method. The generation of intracellular reactive oxygen species (ROS) was detected by using DCFH- DA fluorescent probe. The levels of MDA, SOD and GSH-Px were measured by spectroscopy methods. RESULTS The injury to VERO cells induced by H202 could be reduced obviously by polysaccharide from Cynomorium son- garicum Ruhr. Compared with the model group, polysaccharide from Cynomorium songaricum Ruhr could improve the survival rate of injured cells, reduce the activity of caspase-3 and intracellular ROS ( P 〈 0. 01 ), increase the SOD and GSH-Px activity ( P 〈 0. 01 ) and increase the level of MDA in cell culture. CONCLUSION Polysac- charide from Cynomorium songaricum Ruhr has protective effects on VERO ceils injury induced by H202 in culture and reveals the potential for kidney diseases.
