摘 要:目的建立超高效液相色谱串联质谱(UPLC-MS/MS)方法,检测葛花中异黄酮成分。方法葛花经乙醇提取和乙酸乙酯纯化,HPLC分析采用Diamonsil C18色谱柱(150 mm×4.6 mm,5μm),以甲醇-乙腈-水(2∶1∶2)为流动相,体积流量为0.5 m L/min,检测波长为264 nm,柱温为25℃,进样量10μL,质谱检测设定为负离子模式。结果得到7种异黄酮成分,MS鉴定为大豆苷、大豆苷元、鸢尾苷、鸢尾苷元、染料木黄酮、印度黄檀苷和鸡豆黄素。结论 HPLC-MS/MS法检测葛花异黄酮,具有高灵敏度和选择性,方法准确可靠。AIM To detect isoflavones from the flowers of Pueraria lobata by using ultra-high performance liq- uid chromatography-tandem mass spectrometry (UPLC-MS/MS). METHODS The flowers of Pueraria lobata were extracted by ethanol and re-extracted by ethyl acetate. Analysis of obtained extract was performed on Diamon- sil Cjs (150 mm ×4.6 mm,5μm) column. The mobile phrase was optimized as methanol, acetonitrile, and wa- ter (2 : 1 : 2) at flow rate of 0. 5 mL/min. The detection wavelength was set at 264 nm , the column temperature was maintained at 25 ℃, and the sample volume was 10 μL. The mass spectrograph ran in negative ion mode. RESULTS Seven elements of isoflavones in flowers of Pueraria lobata were identified as daidzin, daidzein, tecto- ridin, tectorigenin, genistein, indian sissotrin and biochanin, respectively. CONCLUSION UPLC-MS detec- ting isoflavone from the flowers of Pueraria lobata has a high sensitivity and selectivity, and the method is accurate and reliable.
