摘 要:目的探讨葛根煨法炮制前后的止泻作用及作用机理。方法将昆明小鼠和SD大鼠分别分为正常组,模型组,阳性对照组,生、煨葛根低、中、高剂量组。除正常组外每组以番泻叶水煎液灌胃,造模成功后分别给予治疗药物,测定小鼠的腹泻指数、胃内残留率和小肠推进率。采用ELISA法检测大鼠血清肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)和白细胞介素10(IL-10),及胃动素(MTL)、胃泌素(GAS)、血管活性肠肽(VIP)的水平。结果与模型组比较,生、煨葛根中、高剂量组均可显著降低腹泻小鼠的腹泻指数(P<0.05),各给药组均能提高腹泻小鼠的胃内残留率,降低小肠推进率(P<0.05);与同剂量的生葛根比较,煨葛根高剂量组小鼠的腹泻指数和胃内残留率有显著性差异(P<0.05);生、煨葛根中、高剂量可降低腹泻大鼠的IL-1β、TNF-α、VIP的水平,升高IL-10、GAS和MTL的水平;与同剂量生葛根比较,煨葛根高剂量组大鼠的IL-10和MTL水平显著升高(P<0.05)。结论葛根能缓解番泻叶引起的腹泻,但煨葛根的止泻作用强于生葛根,其机理可能是通过调节炎性因子来避免肠道的损伤,同时调节胃肠激素分泌使肠道功能趋于正常。AIM To study the anti-diarrhea effect of raw and bran-roasted Puerariae lobatae Radix and their mechanism. METHODS Kunming mice were randomly divided into normal group, model group, positive control group, low, middle and high dosage groups of raw and bran-roasted P. lobatae Radix. All the groups except normal group recieved Sennae Folium, causing diarrhea. Then diarrhea index, gastric residual rate and small intesti- nal propulsion rate were compared among the mice. ELISA was used to measure the contents of rat serum cytokines, including TNF-α, interleukinslβ, interleukin 10, and gastrointestinal hormone, including MTL, GAS, and VIP.RESULTS Middle and high dosage groups of raw and bran-roasted P. Radix could significantly reduce the diarrhea index compared with model group (P 〈 0. 05), each dosage group could improve the gastric residual rate of diarrhea mice, and decrease the rate of small intestine propulsion (P 〈 0. 05), diarrhea index and gastric residual rate in high dosage group had significant difference compared with the same dosage of raw ones (P 〈 0. 05), both raw and roasted ones in middle and high dosage groups reduced the contents of IL-113, TNF-o~ and VIP in diarrhea rats, and elevated the contents of IL-10, GAS and MTL, especially in high dosage group of bran-roasted one significant- ly improved compared with the same dosage of raw one (P 〈 0. 05). CONCLUSION P. lobatae Radix can al- leviate diarrhea caused by Sennae Folium, but high dosage of bran-roasted one is better than the raw one. The anti- diarrheal mechanism may be related to regulation of inflammatory factor in diarrhea to avoid the injury to intestinal, at the same time, harmonize the gastrointestinal hormones to make intestinal function normal.
