摘 要:[目的]观察黄芪六一汤对糖尿病大鼠血糖及胰腺影响。[方法]采取使用随机平行对照方法,将90只SPF级SD大鼠,按随机数字表方法随机分为9组(10只/组);80只大鼠按李大伟方法复制STZ诱导糖尿病大鼠模型,选取血糖居中合格大鼠60只,按随机数字表法随机分6组,模型组、二甲双胍组、消渴丸组、黄芪六一汤(高、中、低剂量)组;造模后第7d分别灌胃干预,黄芪六一汤(生药12.60g/kg,6.30g/kg,3.15g/kg),二甲双胍0.17g/kg,消渴丸0.63g/kg;正常组、模型组同法灌胃等体积生理盐水。连续灌胃干预给药4周后,末次给药前各组大鼠禁食16h,末次给药1h后,常规眼眶静脉丛取血;采血后大鼠脱颈椎处死,取胰腺作病理检查。观测血糖、胰岛素、糖化血红蛋白、细胞形态。[结果]血糖各造模组均高于空白组(P<0.01),血糖值≥13mmolL-1;黄芪六一汤(高、中、低剂量)组血糖均明显低于模型组(P<0.01)。糖化血红蛋白模型组高于空白组(P<0.01),胰岛素含量模型组低于模型组(P<0.01);糖化血红蛋白黄芪六一汤(高、中、低剂量)组明显低于模型组(P<0.01),胰岛素高于模型组(P<0.05)。病理检查空白组胰岛多为圆形或椭圆形细胞团,分散于胰腺腺泡之间,胰岛边界清晰,无包膜,细胞团大小不一,胰岛细胞为多边形,排列整齐,分布均匀,胞浆淡染呈浅粉红色,核多为圆形深染;模型组胰岛细胞可见不同程度肿胀和空泡样变性,边界不清,细胞核大小不等,细胞数量明显减少,排列紊乱,部分细胞发生核固缩和坏死;二甲双胍、消渴丸组胰岛细胞形态结构明显改善,细胞数目增多,排列较整齐,胞浆分布均匀,部分细胞发生肿胀和空泡变性;黄芪六一汤(高、中、低量)组胰岛细胞形态结构明显改善,胰岛细胞肿胀和空泡样变性的程度明显减轻,细胞数目增多,排列较整齐。[结论]黄芪六一汤能显著降低糖尿病大鼠空腹血糖、糖化血红蛋白,升高�[ Objective ] To observe the effects of blood glucose and pancreas of Huangqiliuyi Decoction on experimental diabetic rats. [ Methods ] Taking random parallel control method, 90 SPF SD rats were randomly divided into 9 groups ( 10 / group ) ; 80 rats were Dawei way to copy STZ-induced diabetic rat model, select glucose centering pass 60 rats were randomly divided into 6 groups, model group, metformin group, Xiao Ke Wan group, Huangqiliuyi Decoction group ( high, medium and low dose ) group;Rats were first garage drugs after Modeling 7d, Huangqiliuyi Decoction (crude drug 12.60g/kg, 6.30g/kg, 3.15g/kg), mefformin O.17g/ kg, Xiao Ke Wan 0.63g/kg;Normal group, model group with an equal volume of saline gavage method, after Continuous intragastric administration of four weeks , the rats fasted for 16h before the last administration , after the last administration lh, Blood was taken from the orbital venous plexus;Rats were sacrificed by cervical dislocation after blood collection, Pancreas removed for histological examination. Observation of blood glucose, insulin, glycated hemoglobin, cell morphology. [ Result ] The blood glucose of each model group were higher than control group (P〈0.01), blood glucose≥13mmol·L^-1. The blood glucose of Huangqiliuyi decoction group ( high, medium and low dose ) were significantly lower than the model group (P 〈0.01 ) ;The glycated hemoglobin of model group were higher than the control group ( P 〈0.01 ), the insulin levels of the model group were lower than the control group (P 〈 0.01 );The glycated hemoglobin of Huangqiliuyi decoction group ( high, medium and low dose ) was significantly lower than that in the model group ( P 〈0.01 ), insulin higher than the model group ( P〈0.05 ) .
