摘 要:目的对广东省中山市2007年-2012年间分离的两株W135群脑膜炎奈瑟菌进行病原分子特征分析,调查两者之间的关系,并探索其可能来源。方法使用16S rRNA基因分型、porA可变区分型及多位点序列分型(multilocus sequence typing,MLST)对两株W135群脑膜炎奈瑟菌(ZS07株,ZS12株)进行分子生物学分析。结果两株W135群脑膜炎奈瑟菌分型结果一致,16S rRNA分型为Group 31,MLST型别为ST-11/ET-37(等位基因谱:2,3,4,3,8,4,6),porA测序分型测序为:P1.5,2。结论脑膜炎奈瑟菌ZS07株与ZS12株的亲缘关系高度同源,与我国近几年流行的脑膜炎奈瑟菌亲缘关系较远,与2000年麦加朝圣后由朝觐者引起全球流脑流行的W135群菌株关系密切。Objective To analyze the molecular characteristics of two W135 Neisseria meningitidis isolated from patients in Zhongshan city,Guangdong province,and investigate their relationship and the possible sources. Methods Two W135 Neisseria meningitidis strains( Strain ZS07 and Strain ZS12) isolated from patients in Zhongshan city were examined by three molecular biology approaches: 16 S rRNA gene genotypic,porA gene typing and multilocus sequence typing( MLST). Results These two Neisseria meningitidis strains were characterized in Group 31 by 6S rRNA gene genotypic. The subtype determined by porA variable regions of these two strains was P1. 5,2. The allele profile of the W135 strain in this study was 2,3,4,3,8,4,6,and its sequence type Was ST- 11,which belonged to ST- 1 1 /ET- 37 clone complex. Conclusion The phylogenetic relationship of these two W135 Neisseria meningitides strains was highly homologous. The molecular characteraions of these two W135 Neisseria meningitides strains were different from that of the epidemic strains in China,but they had a close connection with the carriage and transmission of the Hajj epidemic strain in 2000.
