牙龈卟啉单胞菌慢性感染对大鼠颈动脉结构影响

摘　　要：目的探讨牙龈卟啉单胞菌慢性感染对SD大鼠颈动脉结构及其粥样斑块影响。方法将80只10周龄SD大鼠随机分为对照组和牙龈炎组,通过牙颈部丝线结扎并局部接种牙龈卟啉单胞菌的方法建立牙龈炎动物模型,分别在实验第1天(T0)、第1个月(T1)、第3个月(T2)、第6个月(T3)分批处死SD大鼠,取颈动脉,苏木精-伊红(HE)染色进行病理学观察,计算颈动脉横断面内膜与中膜面积的比值(I/M);采用巢式聚合酶链反应(PCR)法检测动脉壁及血液中牙龈卟啉单胞菌16SrDNA。结果 T0、T1时牙龈炎组与对照组I/M值比较差异无显著性(P>0.05),T2、T3时牙龈炎组I/M值大于对照组(F=38.02,q=8.26、10.32,P<0.01)。T1、T2、T3时牙龈炎组血液中检出牙龈卟啉单胞菌16SrDNA,T2、T3时在颈动脉斑块中检测到牙龈卟啉单胞菌16SrDNA。对照组各时期动脉粥样硬化斑块及血液中均未检测到牙龈卟啉单胞菌16SrDNA。结论牙龈卟啉单胞菌慢性感染可加快颈动脉内膜粥样斑块的发展。Objective To investigate the influence of chronic infection of Porphyromonas gingivalis on carotid atherosclerotic plaque in SDrats. Methods Eighty 10-week-old SDrats were equally randomized to control group and periodontitis model group.A periodontitis model was created by ligating dental cervix and local innoculation of Porphyromonas gingivalis.The rats were sacrificed in batches on the first day（T0）,in the first（T1）,third（T2）and sixth（T3）month of the experiment.Carotid arteries were taken for HE staining to do pathologic investigation,to calculate the ratio of intima-media（I/M）thickness of cross section of the carotid,and using nested-polymerase chain reaction（nested-PCR）to analyze P.gingivalis 16 SrDNA. Results The diffe-rences of I/M between the control,T0-and T1-periodontitis groups were not significant,and the I/M value in T2-and T3-periodontitis groups was greater than that in the control（F=38.02;q=8.26,10.32;P〈0.01）.P.gingivalis 16 SrDNA was detected in blood among the periodontitis model groups of T1,T2 and T3,and in plaque of carotid artery of T2 and T3.In the control group,no 16 SrDNA was found in atherosclerotic plaque and blood in each time point. Conclusion Chronic infection of porphyromonas gingivalis may accelerate the progress of carotid atherosclerotic plaque.
