摘  要:目的探讨橙皮素对血管紧张素Ⅱ(Ang Ⅱ)诱导的心肌成纤维细胞增殖及胶原蛋白沉积的影响。方法实验分为空白对照组、Ang Ⅱ组、Ang Ⅱ+橙皮素组、Ang Ⅱ+N-乙酰半胱氨酸(NAC)组,不同因素干预成纤维细胞24h后,采用锥虫蓝染色法检测各组成纤维细胞生存活性的影响;酶标仪检测法检测各组成纤维细胞产生的活性氧(ROS);实时(Real-Time)PCR检测各组成纤维细胞中I、Ⅲ型胶原蛋白及基质金属蛋白酶-1(MMP-1)基因的表达;活细胞计数试剂盒(CCK-8)检测各组成纤维细胞增殖情况。结果橙皮素(25、50、100txmol/L)及AngU(1Ixmol/L)+橙皮素对成纤维细胞生存活性均无影响(均P〉0.05);与空白对照组比,AngII可显著提高成纤维细胞中ROS的表达(1.70±0.12比1,P〈0.01),Ⅰ、Ⅲ型胶原蛋白基因的表达均上调(1.314-0.08、1.404-0.09比1,均P〈0.01),MMP-1基因表达下调(0.68±0.03比1,P〈0.01),AnglI也可诱导成纤维细胞增殖(1.914-0.18比1,P〈0.01)。橙皮素或NAC与Ang Ⅱ同时作用则可逆转这种现象,与Ang Ⅱ组相比,橙皮素(25、50、100Ixmol/L)及NAC(1mmol/L)均可抑制ROS的表达(1.374-0.05、1.16±0.08、1.07±0.07及1.12±0.07比1.70±0.12,均P〈0.01),I、Ⅲ型胶原蛋白基因的表达均下调(1.224-0.08、1.14±0.07、1.02±0.06及1.08.4-0.07比1.31±0.08;1.27±0.07、1.004-0.06、0.99±0.05及1.09±0.06比1.g04-0.09,均P〈0.01),MMP-1基因的表达均上调(0.76±0.05、0.88±0.07、1.01±0.08及0.96±0.07比0.684-0.03,均P〈0.01),细胞的增殖能力也有下降(1.424-0.07、1.384-0.03、1.074-0.15及1.16-t-O.11比1.91.4-0.18,均P〈0.01)。结论橙皮素可抑制Ang Ⅱ诱导的心肌成纤维细胞中ROS的产生,从而抑制胶原蛋白�Objective To explore the effect of hesperetin (HES) on collagen expression in cardiac fibroblasts in vitro induced by angiotensin Ⅱ( Ang Ⅱ). Methods Cell growth was determined by trypan blue staining, reactive oxygen species (ROS) generation by microplate reader, the expressions of collagen I , III and matrix metalloproteinase 1 (MMP-1) by real-time polymerase chain reaction (PCR) and cell proliferation by cell counting kit-8. Results HES and Ang Ⅱ + HES had no effect on cellular activity. Ang Ⅱ significantly increased ROS generation ( 1.70 ± 0. 12 vs 1, P 〈 0. 01 ) , gene expression of collagen I , m both increased (1.31 s0. 08 vs 1, 1.40 s0.09 vs 1, both P 〈0.01 ) while MMP-1 decreased (0. 68 ± 0.03 vs 1, P〈0.01). Ang Ⅱ also induced the proliferation of fibroblasts (1.91 ±0. 18 vs 1, P〈0.01). While HES (25, 50, 100 p±mol/L) or NAC (1 retool/L) reversed these effects during co-treating with Ang l], ROS decreased versus the Ang II group(l. 37 ±0.05,1.16 ±0.08,1.07 ±0.07,1.12 ±0.07 vs 1.70 ± 0. 12, all P 〈 0. 01 ) , gene expression of collagen I , m also decreased ( 1.22 ± 0.08 and 1.27 ± 0.07, 1.14±0.07 and 1.00 ±0.06, 1.02 ±0.06 and 0.99 ±0.05, 1.08 ±0.07 and 1.09 ±0.06 vs 1.31 ±0.08 and 1.40 ±0.09, all P〈0.01), MMP-1 increased (0.76 ±0.05, 0.88 ±0.07, 1.01 ± 0.08, 0. 96 ± 0.07 vs 0. 68 ± 0.03, P 〈 0. 01 ) versus the Ang lI group. Cell proliferation was alsoinhibited (1.42±0.07, 1.38+0.03, 1.07+0.15, 1.16±0.11 vsl. 91±0.18, allP〈0.01). NAChad the same effect with HES. Conclusion HES inhibits the synthesis of collagen and the Ang Ⅱ-induced proliferation of fibroblast through suppressing the ROS generation.
