摘 要:目的 探讨微小RNA (miR)-17-5p在特发性少精症患者睾丸组织中的表达及其意义.方法 对25例特发性少精症患者和10例正常男性为对照组,分别采用实时定量聚合酶链反应(Real-time PCR)和Western blot检测miR-17-5p基因和E2F1蛋白在其睾丸组织中的表达,应用原位缺口末端标记法(TUNEL)检测睾丸组织切片中生精细胞凋亡率.结果 Real-time PCR显示miR-17-5p基因表达在特发性少精症患者睾丸组织中较正常生育能力的睾丸组织显著减少(0.38),明显低于对照组(1.00,P<0.05),Western blot结果显示特发性少精症患者组睾丸组织中凋亡蛋白E2F1条带灰度值显著高于正常对照组(P<0.05),凋亡率检测结果示特发性少精症组凋亡指数(AI)为(50.8±0.9)%,显著低于正常对照组的(1.3±1.2)%(P<0.05).结论 miR-17-5p表达减少可能通过上调凋亡蛋白E2F1表达,而促进睾丸生精细胞过度凋亡,导致特发性少精.Objective To investigate the mRNA expression of microRNA (miR)-17-5p in the testis tissue of patients with idiopathic oligozoospermia.Methods The testis tissues in 25 cases of idiopathic oligozoospermia were examined by real-time quantitative polymerase chain reaction(Real-time PCR) for detection of the miR-17-5p expression.Western blotting was used to detect the E2F1 protein and TdT-mediated dUTP nick end labeling (TUNEL) to measure cell apoptosis.Results The expression of miR-17-5p was significantly lower in testicular tissues of patients with idiopathic oligozoospermia than that in normal testis tissues (0.38 vs.1.00,P 〈0.05).However,the expression of E2F1 protein in testicular tissues of patients with idiopathic oligozoospermia was significantly higher than that in normal testis tissues,and the apoptosis rate of patients with idiopathic oligozoospermia was significantly higher [apoptosis index (AI),(50.8±0.9)% vs.(1.3 ± 1.2)%,P〈0.05].Conclusion The aberrant decreased expression of miR-17-5p may contribute to idiopathic oligozoospermia through promoting cell apoptosis.
