摘 要:目的:在动脉粥样硬化模型鼠中,观察白藜芦醇对主动脉内粥样硬化病变及斑块中氧化型低密度脂蛋白的受体:植物血凝素样氧化型低密度脂蛋白受体1(lectin-like oxidized low density lipoprotein receptor-1,LOX-1)及基质金属蛋白酶9(Matrix metal oproteinase-9, MMP-9)表达的影响。方法:雄性C57BL/6J小鼠32只及雄性载脂蛋白E基因缺陷(apo E-/-)小鼠32只,均6周龄,随机均分为4组:普通饲料组、普通饲料+白藜芦醇组、高脂饲料组、高脂饲料+白藜芦醇组,每组均8只。喂养至11月龄后行主动脉根部油红O染色,测量主动脉窦动脉粥样硬化斑块负荷。行免疫组织化学染色,观察主动脉斑块内LOX-1及MMP-9蛋白的表达。结果:C57BL/6J 小鼠各组均未见动脉粥样硬化及明显的 LOX-1及MMP-9蛋白表达。apo E-/-小鼠在11月龄出现明显主动脉粥样硬化,高脂组较主动脉内动脉粥样硬化斑块体积及 LOX-1、MMP-9表达均明显高于普通饲料组(P 〈0.05)。白藜芦醇对动脉粥样硬化斑块体积及LOX-1、MMP-9表达均无显著影响。结论:高脂饮食可促进动脉粥样硬化模型小鼠动脉内动脉粥样硬化的形成及LOX-1、MMP-9蛋白的表达,白藜芦醇对此无显著影响。This was planned to study the effect of resveratrol on the aorta atherosclerotic on artery atherosclerotic plaque formation and on the expression of lectin-like oxidized LDL receptor-1(LOX-1) and matrix metal oproteinase-9 (MMP-9) in the elderly apolipoprotein E knockout (apo E-/-) mice model. Thirty-two male apo E-/-mice,with thirty-two male C57BL/6J mice as controls, al 6 weeks,were randomized into four groups:normal diet group ,normal diet with resveratrol group, high-fat diet group, and high-fat diet with resveratrol group . At 11 month, mice of each group were kil ed. The expressions of LOX-1 and MMP-9 in aortic wal were studied by immunohistochemistry. The burden of atherosclerotic plaque of aorta root was measured by oil red O staining. None of C57BL/6J mice were observed with obvious LOX-1 and MMP-9 protein expression or with atherosclerotic plaque. As compared with the normal diet groups, high-fat diet significantly increased the expressions of LOX-1and MMP-9 and also the burden of atherosclerotic plaque in aorta in apo E-/-mice(P〈0.05). Resveratrol had no effect on artery atherosclerotic plaque formation and on the expression of LOX-1 and MMP-9 in the elderly Apo E-/-mouse. Resveratrol had no effect on artery atherosclerotic plaque formation and on the expression of LOX-1 and MMP-9, which could be increased by high-fat diet, in the elderly Apo E-/-mouse.
