摘  要:目的:观察牛蒡子苷元对S180荷瘤小鼠肿瘤组织生长及其血管生成的影响。方法:以S180荷瘤小鼠为研究对象,设置阴性对照组(溶剂)、阳性药物组5-氟尿嘧啶40mg/kg和牛蒡子苷元低、中、高剂量组(40、80、160 mg/kg),按0.01 ml/g,每日灌胃给药1次,连续10 d,脱颈椎处死小鼠,对肿瘤组织及脾脏称重并计算抑瘤率及脾脏指数,用免疫荧光染色法检测肿瘤组织内微血管密度(MVD)和血管生长因子VEGF的表达。结果:牛蒡子苷元不同剂量组对S180荷瘤小鼠肿瘤的生长和肿瘤组织中的微血管密度(MVD)及VEGF的表达均有显著的抑制作用,牛蒡子苷元40、80、160 mg/kg的抑瘤率分别为23.00±13.10%,40.42±12.90%和45.67±16.15%,肿瘤组织微血管密度(MVD)分别为38.25±4.53、32.62±3.66和22.75±5.28,免疫组化染色VEGF相对光密度值分别为75.66±8.41%、70.32±5.73%和60.66±9.38%。结论:牛蒡子苷元能够抑制S180荷瘤小鼠肿瘤生长及其瘤内微血管密度,对肿瘤组织内VEGF的表达亦有抑制作用。To observe the anti-angiogenesis effect of arctigenin in tumor tissue of tumor-bearing mice. Method: The S180 tumor-bear- ing mice was used to detect the anti-angiogenetic effect of arctigenin in vivo. Divide the tumor-bearing mice into five group: control group ( blank solvent), 5-fluorouracil (40 mg/kg), aretigenin low, medium, high dose group ( 40,80,160 mg/kg). Each group was continuously given corresponding medicine ten days by gavage, and then the mice were sacrificed by cervical dislocation. Weigh the tumor tissue and spleen, calculated inhibition rate and spleen index. The expression of tumor microvessel density (MVD) and vascular endothelial growth fac- tor (VEGF) was detected by immunohistochemical staining. Result: Tumor growth and tumor tissue mlcrovessel density (MVD) and VEGF expression was significantly inhibited by arctigeninin in a dose-dependent manner. When the concentrations of rctigenin were 40 mg/kg,80 mg/kg,160 mg/kg,the inhibition rates on tumor were 23.00 ± 13.10% ,40.42 ± 12.90% and 45.67 ± 16.15% ,the microvessel density were 38.25 ± 4.53,32.62 ± 3.66 and 22.75 ± 5.28, the protein expression of VEGF were 75.66 ± 8.41% ,70.32 ± 5.73 % and 60.66 ±9. 38%. Conclusion: Arefigenin earl inhibit tumor growth and the mierovessel density of S180 tumor-bearing mice,the expression of VEGF was also inhibited.
