The Influence of Different Preconditioning Methods on the Expression of Myocardial Cell Adhesion Molecules in Overload Training Rats

摘  要:目的:观察预热处理、药物预处理和穴位电刺激预处理对超负荷训练后大鼠心肌细胞粘附因子表达的影响,比较三种预处理方法对超负荷训练后心脏缺血局部炎性反应的作用。方法:25只雄性SD大鼠随机均分为A组(对照组)、B组(训练组)、C组(预热处理组)、D组(药物预处理组)和E组(穴位电刺激预处理组),后3组在进行耐力训练8周的同时分别再予以高热、川穹嗪和穴位电刺激预处理。采用免疫组化法观测心肌组织中细胞间粘附分子-1(ICAM-1)、血管细胞粘附分子-1(VCAM-1)和血小板内皮细胞粘附分子-1(PECAM-1)表达。计算大鼠心肌细胞ICAM-1、VCAM-1、PECAM-1阳性区域平均光密度(MOD)。结果:超负荷训练可使大鼠心肌中ICAM-1、VCAM-1、PECAM-1表达明显增高,经过适当的温度、药物和穴位电刺激预处理后ICAM-1、VCAM-1、PECAM-1表达显著下降。B、C、D、E 4组大鼠心肌细胞ICAM-1、VCAM-1、PECAM-1阳性区域MOD高于A组,差异有统计学意义(P<0.01),B组与C、D、E组比较也存在显著性差异(P<0.05),C、D、E 3组之间比较未见显著性差异(P>0.05)。结论:穴位电刺激预处理与预热处理、药物预处理均可显著抑制超负荷训练后心肌ICAM-1、VCAM-1和PECAM-1高表达,减轻损伤心肌局部的炎症反应,提示对损伤心肌局部炎性反应的抑制可能也是预处理预防超负荷训练后心肌损伤的机制之一。Objective To validate the efficacy of heat preconditioning(HP),drug(Ligustrazine)preconditioning and acupuncture preconditioning in reducing local inflammatory reaction in myocardium after overload training through the changes in expression of myocardial cell adhesion molecules.Methods Twenty five male SD rats were randomly and equally divided into following five groups:group A(control group),group B(8-week overload training group),group C(8-week HP before overload training),group D(8-week drug preconditioning before overload training)and group E(8-week electrical acupoint stimulation on Neiguan and Zusanli before overload training). The expressions of myocardial intercellular adhesion molecule-1(ICAM-1),vascular cell adhesion molecule-1(VCAM-1),and platelet-endothelial cell adhesion molecule-1(PECAM-1)were observed by immunohistochemical staining. Results Expressions of ICAM-1, VCAM-1 and PECAM-1 in myocardium of rats increased significantly after overload training, whereas decreased significantly after appropriate heat, drug and acupuncture preconditioning. The mean optical density(MOD)of ICAM-1,VCAM-1,and PECAM-1 in myocardium of groups B,C,D and E were significantly higher than that of group A(P 〈0.01),and of group B was significantly different from groups C,D and E(P〈 0.05). There was no significant difference in MOD among groups C,D and E(P〉 0.05). Conclusion Preconditioning via electrical acupoint stimulation,appropriate heat preconditioning and drug preconditioning could significantly inhibit the expression of ICAM-1,VCAM-1 and PECAM-1 in myocardium after overload training,and thus reducing local inflammatory and preventing myocardial injury.
