Effects of Exhaustive Exercise and Contusion Injury on Satellite Cells and Related Regulatory Factors in Skeletal Muscle of Rats

摘  要:目的:研究力竭运动及钝挫伤对大鼠骨骼肌卫星细胞及血小板衍生生长因子(PDGF)和转化生长因子β(TGF-β)的影响。方法:力竭运动组:7周龄Wistar雄性大鼠24只,随机分为3组,每组6只,进行一次性力竭跑台运动,分别于力竭后即刻、24小时、48小时宰杀(即E0、E24、E48组),并取左侧腓肠肌一分为二,一份立即进行骨骼肌卫星细胞培养实验,另一份液氮保存,用于检测血小板衍生生长因子(PDGF)和转化生长因子β(TGF-β)的变化,采用ELISA法。对照组(C组):不运动,与EO组同时宰杀。钝挫伤组:Wistar雄性大鼠4只,进行钝挫伤实验,于钝挫伤后即刻、24小时(D0、D24组)分别进行麻醉,取左侧腓肠肌,每组2只,立即进行细胞培养实验。另取新生鼠(N组)2只,宰杀后取左侧腓肠肌,同样进行细胞培养实验。结果:(1)体外细胞培养显示,新生鼠(N组)细胞培养1天后即可见到梭形卫星细胞,钝挫伤即刻组(D0组)3天后可见梭形卫星细胞,而钝挫伤后24小时组(D24组)5天后可见梭形卫星细胞,且此三组分化的卫星细胞一直保持良好的增殖、传代,第13天仍长势良好,可见部分融合和微管。另外,本实验条件下,对照组(C组)和力竭运动组(E0、E24、E48组)未能观察到明显的卫星细胞增殖、分化。(2)PDGF和TGF-β的ELISA结果显示,一次力竭离心运动后,与安静对照组相比较,力竭运动组大鼠骨骼肌中PDGF含量均显著性升高(P<0.01),而TGF-β含量均呈现极显著性降低(P<0.01)。且力竭运动各组(E0、E24、E48组)之间无显著性差异(P>0.05)。结论:钝挫伤刺激对骨骼肌卫星细胞有较强的激活作用,PDGF在肌肉损伤修复中可能具有正性作用,而TGF-β则可能起着负性调节因子的作用。Objective The paper investigated how exhaustive exercise and contusion injury affected satellite cells,platelet growth factor(PDGF) and transforming growth factor-beta(TGF-beta) in skeletal muscle of rats. Methods 24 Wistar male rats(7 weeks old)were randomly divided into four groups(n =6): the control group(C),immediately after exhaustive exercise group(E0),24 hours after exhaustive exercise group(E24),48 hours after exhaustive exercise group(E48). The rats in all exercise groups were forced to perform acute treadmill exhaustive exercise and killed immediately,24 hours,and 48 hours after exhaustive exercise. Then the left gastrocnemius muscle were removed,part of the muscle was used for cell culture in vitro immediately,and the other(stored in liquid nitrogen)for measuring PDGF and TGF-beta by ELISA. At the same time,contusion of 4 Wistar male rats(7 weeks old)were established according to Kami et al. Their left gastrocnemius was removed immediately after contusion injury(D0,n= 2)and 24 hours after contusion(D24,n = 2)for the satellite cell culture in vitro. Additionally,gastrocnemius muscle from two newborn rats was also removed and underwent cell culture immediately.Results(1)A small amount of spindle satellite cells appeared in the newborn rats one day after culture,whereas 3 days in group D0 and 5 days in D24 group after culture. They kept good proliferation until13 thday after serial passage,in which fusion and microtubule partially revealed. On the other hand,no proliferation and differentiation of satellite cells was observed in groups C,E0,E24,and E48 under the same culture condition.(2)There were significant increase in PDGF content and decrease in TGF-βcontent in skeletal muscle in groups E0,E24 and E48,as compared with the group C(P〈 0.01).
