仙灵脾益源胶囊增加骨密度的原料配伍合理性实验研究

摘　　要：目的:观察仙灵脾益源胶囊不同组成原料的配伍对维甲酸大鼠骨质疏松模型的影响,为中药保健食品原料配伍合理性提供实验依据。方法:雌性Wistar大鼠随机分为空白组、模型组、全方组(A+B+C)、中药组(A)、天然产物组(B)、营养成分组(C)、A+B组、A+C组、B+C组。采用70 mg·kg-1维甲酸灌胃法建立大鼠骨质疏松模型,造模同时开始灌胃给药,连续4周。给药结束后,左侧股骨经骨密度仪测量骨密度;右侧股骨置105℃烘箱中16 h至质量恒定,游标卡尺测量股骨中点直径,原子吸收法测定骨钙含量;放免法测定血清中骨钙素水平。结果:与空白组比较,模型组大鼠骨密度与股骨中点直径显著降低(P<0.05),血清骨钙素水平显著升高(P<0.05),股骨钙含量显著降低(P<0.01)。与模型组比较,A+B+C全方组可显著升高大鼠骨密度(P<0.05),降低血清骨钙素水平(P<0.05),提高股骨骨钙含量(P<0.01);C组可显著降低血清骨钙素水平(P<0.05),A+B组和A+C组则可显著升高大鼠股骨骨钙含量(P<0.01,P<0.05)。与A+B+C全方组比较,A组股骨中点直径增加明显,B+C组、B组、C组股骨骨钙含量显著降低(P<0.05)。结论:仙灵脾益源胶囊能够改善骨质疏松大鼠的骨密度、降低骨转换水平及增加骨钙含量,其原料两两配伍或单独应用在各指标虽有一定作用趋势,但均不如三者联合配伍作用显著。Objective： To study the rationality of Xianlingpi Yiyuan Capsule and its components and its impact on osteoporosis. Methods：Female Wistar rats were randomly divided into sham-operate group,model group,the whole party group（ A ＋ B ＋ C）,Chinese medicine group（ A）,natural products group（ B）,nutrition group（ C）,A ＋ B,A ＋ C group,B ＋ C group. Rat osteoporosis models was established by intragastric administration of retinoic acid（ 70 mg·kg^-1） for 2 weeks. Rats in the day gavage for consecutive 4 weeks. After medication,take the left femur for determination the bone mineral density（ BMD）,take the right femur to 105 ℃ oven 16 hours for constant quality,and take the serum for detemination the osteocalcin. The femur middle point diameter was measure by vernier caliper. Results：Compared with the sham-operate group,values of BMD and femoral middle point diameter significantly decreased（ P〈0. 05）,the serum osteocalcin level obviously increased（ P〈0. 05）,femoral calcium content decreased significantly（ P〈0. 01）. Compared with model group,A ＋ B ＋ C all party group can significantly increase the BMD of rats（ P〈0. 05）,decrease serum osteocalcin level（ P〈0. 05）,increase the femoral bone calcium content（ P〈0. 01）; C can significantly reduce serum osteocalcin level（ P〈0. 05）,A ＋ B,A ＋ C group can improve model rats femur bone calcium content（ P〈0. 01,P〈0. 05）. Compared with A ＋ B ＋ C the whole party group,middle diameter significantly increased in A,and B ＋ C group,B group and C group femoral bone calcium content decreased significantly（ P〈0.05）. Conclusion： Xianlingpi Yiyuan Capsule can improve the osteoporosis rats BMD,and decrease the levels of bone turnover and increase bone calcium content,the effect of all party group was better than one material or double materials.
