常山酮对人急性单核细胞白血病细胞增殖、凋亡及相关基因表达的影响

摘　　要：目的:研究常山酮对人急性单核细胞白血病(THP-1)细胞增殖、凋亡及Bax、Bcl-2 mRNA表达的影响。方法:取对数生长期的THP-1细胞,分别加入不同浓度的常山酮,CCK-8法检测THP-1增殖率;流式细胞术检测THP-1凋亡情况;实时定量PCR检测Bax及Bcl-2 mRNA的表达情况。结果:常山酮能抑制THP-1细胞增殖、诱导其凋亡,呈剂量依赖关系。常山酮作用后,THP-1细胞内Bax mRNA表达显著升高,而Bcl-2 mRNA表达显著下降。结论:常山酮能抑制THP-1细胞增殖、诱导其凋亡,其作用机制可能与上调Bax mRNA表达、下调Bcl-2 mRNA表达相关。Objective：To investigate the effect of halofuginone on apoptosis and Bcl-2/Bax mRNA level of THP-1 cells.Methods：THP-1 cells were cultured with different concentration of halofuginone,cell proliferation was detected by CCK-8;cell apoptosis were examined by flow cytometry;and the level of Bcl-2/Bax mRNA was detected by real-time PCR.Results：Halofuginone could inhibit proliferation and promote apoptosis of THP-1 cells in a dose-dependent manner.After treated with halofuginone,the level of Bax mRNA in THP-1 cells was significantly increased,whereas the level of Bcl-2 mRNA was remarkably decreased.Conclusion：Halofuginone could inhibit proliferation and promote apoptosis of THP-1 cells,which might partly be through down-regulating the level of Bcl-2 mRNA and up-regulating the level of Bax mRNA.
