基质金属蛋白酶及其抑制剂对肺纤维化大鼠MMP-2和TIMP-1表达的影响

摘　　要：目的研究红景天苷(SDS)对百草枯(PQ)中毒大鼠肺纤维化肺组织中基质金属蛋白酶-2(MMP-2)及其组织特异性抑制因子-1(TIMP-1)表达的影响。方法 SD大鼠随机分为对照组,PQ模型组和SDS治疗组。一次性灌胃PQ制备肺纤维化模型,治疗组腹腔注射SDS(10 mg/kg,q12 h);正常对照组腹腔注射等体积NS。PQ模型组和SDS治疗组分别在染毒后相应时间点(1 d,3 d,7 d,14 d,21 d,28 d)麻醉处死大鼠;取其肺组织,进行HE染色;免疫组化和Real-Time PCR检测MMP-2、TIMP-1蛋白与mRNA表达。结果与NS组比较,模型组MMP-2 mRNA及其蛋白、TIMP-1 mRNA及蛋白的表达明显增强(P<0.01);各干预组肺组织表达相对减低(P<0.05)。结论 SDS能降低MMP-2及TIMP-1的mRNA及蛋白的表达,抑制PQ中毒大鼠肺组织的纤维化。Objective To observe the effects of salidroside（ SDS） and paraquat（ PQ） on the expression of matrix metalloproteinase（ MMP- 2） and tissue inhibitor of metalloproteinase- 1（ TIMP- 1） in paraquat poisoning pulmonary fibrosis rats.Methods SD rats were randomly divided into three groups,normal control group（ NS）,paraquat（ PQ） model group and salidroside（ SDS） intervention group. The pulmonary fibrosis model induced by PQ poisoning was given a gavage of PQ solution,rats in SDS intervention group intraperitoneal injection SDS solution,and normal control group（ NS） were given normal saline solution with the same volumeand. The normal control group directly executed rats after anesthesia,PQ model group and SDS intervention group at 1 d,3 d,7 d,14 d,21 d,28 d the rats were sacrificed,and the lungs were taken away for HE staining detecting lung tissue pathological changes; Immunohistochemical method was used to detect MMP- 2 and TIMP- 1 protein expression; Real- Time PCR to detect the expression of MMP- 2 and TIMP- 1 mRNA. Results Compared with NS group,PQ model groups the mRNA and protein expression of MMP- 2 was obviously increased（ P〈 0 01）; RNA and protein expression of TIMP- 1was obviously increased（ P〈 0 01）. Compared with PQ model group,SDS intervention group at each time point lung tissue MMP- 2 and TIMP- 1 protein expression significantly reduced,there is significant difference（ P〈 0 05）. Conclusion SDS can decrease the expression of MMP- 2 and TIMP- 1 mRNA and protein,inhibition of fibrosis in the lung tissue of rats PQ poisoning.
