摘  要:目的:合成新型的功能性氧化石墨烯-β-环糊精(GO-DEX-β-CD)药物载体和功能性氧化石墨烯-β-环糊精/利多卡因(GO-DEX-β-CD/LDH)包合物,并对其进行质量评价。方法:利用电子扫描显微镜(SEM)、原子力显微镜(AFM)对其形貌进行表征;以高效液相色谱法(HPLC)测定利多卡因(LDH)含量,并对生成物的包封率、回收率以及精密度进行评价,以离心法测定GO-DEX-β-CD/LDH包合物的包封率。结果:GO-DEX-β-CD/LDH包合物的平均粒径(n=15)为(121±5.23)nm,LDH检测浓度线性范围为20~100μg·mL-1(r=0.9990),平均回收率(n=3)为(100.4±0.44)%,包封率(n=5)为(72.1±1.12)%,GO-DEX-β-CD/LDH包合物具有良好的稳定性,对皮肤无刺激性。结论:GO-DEX-β-CD/LDH包合物制备工艺可行,质量控制方法简便可靠。Objective :To synthetize the new- type GO -DEX-β- CD cartier and the GO -DEX -β- CD/LDH inclusion compound,and evaluate their qualities. Methods: The microstructure of graphene oxid was characterized by scanning electron microscopy (SEM) and atomic force microscopy (AFM). The concentration of lidocaine (LDH) was determined by high efficiency liquid chromatography. The recovery, precision and entrapment efficiency of GO- DEX-β- CD/LDH inclusion compound were studied,and the GO- DEX-β- CD/LDH inclusion com- pound entrapment efficiency was determined by centrifugation. Results: The mean diameter (n = 15 ) of GO - DEX -β - CD/LDH inclusion compound was ( 121 ± 5.23 )nm, the linear detection concentration range of LDH was 20 -100 μg · mL-1( r = 0. 9990) with the average recovery (n = 3 ) of (100.4± 0.44)% and the entrapment effi- ciency (n = 5 ) of (72. 1± 1. 12)%. GO - DEX -β - CD/LDH inclusion compound had a good stability and showed no skin irritation. Conclusion: The preparation technique of GO -DEX -β- CD/LDH inclusion compound is feasible, and the quality control method is convenient and reliable.
