补气生血系列药对含药血清对血虚小鼠骨髓造血功能的影响

摘　　要：目的:观察补气生药系列药对含药血清对血虚模型小鼠骨髓造血功能的影响,为阐明中医"补气生血"治则提供实验依据。方法:连续4天腹腔注射环磷酰胺30 mg/kg复制化学性血虚小鼠模型;灌胃7.5 g生药/kg各补气生血药对水煎液,一日两次,第4天腹主动脉取血,分离含药血清;观察各组含药血清对骨髓造血祖细胞CFU-GM、CFU-E、CFU-B的影响及骨髓有核细胞DNA合成的影响;建立定性均匀设计中虚拟变量的多元线性回归方程,找出最佳药效配伍;运用双因素方差分析补气药与补血药的贡献度。结果:补气生血各药对的含药血清均能提高小鼠骨髓CFU-GM、CFU-E、BFU-E和OD268值;通过主成分分析得到的多元回归方程为:YF=-0.771+2.226人参+1.351黄芪+1.169西洋参+1.082白芍+0.706当归+0.464熟地;在提高CFU-GM和骨髓有核细胞DNA合成方面,补气药与补血药有交互作用。结论:补气生血各药对含药血清可能够促进骨髓粒系、红系造血祖细胞的增殖与分化而促进骨髓造血;对于骨髓造血功能作用最强的是人参白芍药对;补气药可增加补血药的药效而达到其"补气生血"功效。Objective ： By observing the effect of serums containing Reinforcing Qi and Engendering Blood Prescription on bone marrow hematopoietic function in blood deficiency mice to provide experimental data for rule of invigorating qi and promoting blood. Methods：Chemical blood deficiency model was replicated with intraperitioneal injection of cyclophosphamide ,30 mg/kg, l0 mL/kg, once a day for 4 days. 7.5 g/kg Reinforcing Qi and Engendering Blood Prescription was administrated intragastrically twice a day, and serums containing Engendering Blood Prescription were extracted by abdominal aortic blood on the 4th day. The bone marrow nucleated cells of blood - deficient mice were cultured in vitro, and the bone marrow CFU - GM, BFU - E and DNA content were observed. Multiple liner regression with dummy variable was set up to find out the best efficient prescription. Two - factor anova was used to analyze the contribution of supplying drugs and blood tonic. Results ： The number of bone marrow CFU - GM colony, CFU - E colony, BFU - E colony, and the content of OD26s can be improved by serums containing Reinforcing Qi and Engendering Blood Prescription. Qi -reinforcing medicine and blood -enriching medicine have interactions on improving CFU - GM colony and marrow nucleated cell DNA synthesis. Conclusion：Serum containing Reinforcing Qi and Engendering Blood Prescription could promote marrow hemopoiesis by increasing the marrow hematopoietic progenitor cells multiplication and differentiation. The best compatibility was Ginseng Radix et Rhizoma and Paeoniae Radix Alba. Qi - reinforcing medicine can increase the efficacy of hematopoietic and reaches its effect of invigorating Qi and promoting blood.
