摘  要:[目的]研究黄芪甲苷对肝缺血/再灌注损伤的保护作用及其机制。[方法]将雄性SD大鼠36只,随机分为3组:假手术组(Sham组)、肝缺血/再灌注组(I/R组)、黄芪甲苷预处理组(AsIV组)。分光光度法测定大鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性;苏木精-伊红(HE)染色观察肝组织形态学变化,蛋白印迹法(Western blot)检测心肌组织内Bax,Bcl-2蛋白表达。[结果]与假手术组比较,肝I/R组ALT、AST活性显著增高,Bax蛋白表达增加,Bcl-2蛋白表达降低,黄芪甲苷预处理可以降低肝缺血/再灌注损伤引起的ALT、AST活性增高,Bax蛋白表达增加,增加Bcl-2蛋白表达。[结论]黄芪甲苷预处理对肝缺血/再灌注损伤有一定的保护作用,其机制可能与黄芪甲苷抑制肝细胞凋亡有关。[Objective] To investigate the protective effect of Astragaloside IV (As IV) against liver ischemia and reperfusion injury and the underlying mechanism. [Methods] The 36 SD rats were randomly divided into 3 groups: sham operation group (Sham group), liver ischemia-reperfusion (I/R group) group and As IV group. The content of ALT and AST were measured by spectrophotometric method. Liver pathological changes were observed by HE staining. Bax and Bcl-2 protein expression was determined by Western blot. [Results] Compared with sham group, the content of ALT and AST was significantly increased, while the Bcl-2 protein expression decreased and the Bax expression increased. Pretreatment of As IV before liver I/R could decrease the content of ALT and AST, Bax expression, and increase the Bcl-2 protein expression (P〈0.05). [Conclusion] As IV pretreatment has a protective effect against liver ischemia reperfusion injury, which is partially finished via inhibiting liver cell apoptosis.
