摘 要:目的观察阳和平喘颗粒对哮喘大鼠气道黏液高分泌的干预作用,探究其作用机制。方法将50只清洁级SD大鼠随机分为正常组、模型组、阳和平喘颗粒低剂量组、阳和平喘颗粒高剂量组和地塞米松组,每组10只。采用卵清蛋白成功诱发大鼠哮喘后,并予相应药物干预8周。阿尔辛蓝-过碘酸雪夫氏染色,光镜下观察气道杯状细胞;采用免疫组织化学法检测大鼠气道黏蛋白5AC(mucin 5AC,MUC5AC)的表达水平;采用Western blot、RT-PCR分别检测大鼠肺组织转化生长因子β1(transforming growth factor beta 1,TGF-β1)蛋白及其mRNA表达水平。结果阳和平喘颗粒高、低剂量组及地塞米松组大鼠气管可见少量黏液分泌,未见明显杯状细胞增生;与模型组比较,阳和平喘颗粒高、低剂量组及地塞米松组大鼠气道组织杯状细胞面积和MUC5AC蛋白表达水平,以及肺组织TGF-β1蛋白及TGF-β1 mRNA表达水平均显著降低(P<0.01);阳和平喘颗粒高剂量的效应显著优于低剂量,呈现明显的剂量依赖性(P<0.01)。结论阳和平喘颗粒可降低哮喘大鼠气道黏液高分泌,机制可能与其下调肺组织TGF-β1蛋白及其基因表达水平,抑制MUC5AC生成有关。Objective To observe the effects of Yanghe Pingchuan Granule on the airway mucus hypersecretion in asthmatic rats and to investigate its action mechanism.Methods Fifty clean Sprague-Dawley rats were randomly and equally divided into normal group,model group,low-and high-dose Yanghe Pingchuan Granule groups,and dexamethasone group.Asthma was induced with ovalbumin in rats,which were then given medication for 8 weeks.Alcian blue-periodic acid Schiff staining was performed to observe airway goblet cells.The expression of mucin 5AC (MUC5AC) in the airway was measured by immunohistochemistry.The mRNA and protein expression of transforming growth factor-beta 1 (TGF-β1) in rat lung tissue was measured by RT-PCR and Western blot,respectively.Results The low-and high-dose Yanghe Pingchuan Granule groups and dexamethasone group showed little mucus secretion and no proliferation of goblet cells.Compared with the model group,the low-and high-dose Yanghe Pingchuan Granule groups and dexamethasone group showed significant decreases in the area of goblet cells in airway tissue,MUC5AC protein expression,and the mRNA and protein expression of TGF-β1 in lung tissue (P<0.01).Yanghe Pingchuan Granule had significantly better efficacy at a high dose than at a low dose,suggesting a significant dose dependence (P<0.01).Conclusion Yanghe Pingchuan Granule can reduce airway mucus hypersecretion in asthmatic rats,possibly by downregulating the mRNA and protein expression of TGF-β1in lung tissue and inhibiting the formation of MUC5AC.
