摘 要:目的:观察化瘀消癥杀胚中药对异位妊娠裸鼠体内移植模型的影响。方法:通过绒毛组织块移植法建立人源妊娠绒毛组织块裸鼠体内移植模型,中药灌服7d,并设西药组(肌注甲氨蝶呤)和阴性组(灌服纯净水)。观察裸鼠一般生长情况、包块情况,给药前后测裸鼠绒毛膜促性腺激素(CG),采用qRT-PCR检测种植包块的雌、孕激素受体的表达。结果:裸鼠一般生长情况良好,3组包块变化、CG变化比较差异无统计学意义。中药组和西药组雌激素受体(ER)、孕激素受体(PR)表达量较阴性组均明显降低,差异均有统计学意义(P<0.05),中药组与西药组的ER、PR表达量比较差异无统计学意义。结论:化瘀消癥杀胚中药能够下调ER、PR表达,从受体水平拮抗雌孕激素,从而导致绒毛组织因缺乏雌孕激素而变性、坏死。Objective: To observe the effects of Huayu Xiaozheng Shapei Decoction on ectopic pregnancy nude mice model. Methods: The experimental nude mice were randomly divided into Chinese medicine (Huayu Xiaozheng Shapei Decoction) group, western medicine (MTX) group and model group. The human chorionic tissue xenograft nude mice model was made by human chorionic tissue transplant method. The Chinese medicine group was gavaged with Huayu Xiaozheng Shapei Decoction for succesive 7 days, while the model group was gavaged with purified water in the same volume and the western medicine group was intramuscular injected with MTX. The general situation and plant mass of each group were observed and the CG level were measured before and after the treatment. The expression of estrogen receptor (ER) and progesterone receptor (PR) of plant mass were detected by qRT-PCR technology. Results: There was no significant difference in general situation, plant mass and CG level among these three groups. The ER and PR expression in Chinese medicine group and western medicine group were both significantly lower than those in model group, with statistical differences (P〈0.05). There was no significant difference in the ER and PR expression between Chinese medicine group and western medicine group. Conclusion: Huayu Xiaozheng Shapei Decoction could down-regulate the expression of ER and PR, the lack of which will result in the degeneration and necrosis of chorionic tissue, which may be its molecular mechanism.
