摘  要:目的:探讨电针内关穴对异丙肾上腺激素(ISO)诱导的心肌缺血大鼠心肌氯离子通道CLC-3、CLC-4蛋白表达的影响。方法:使用皮下注射IOS的方法制备大鼠心肌缺血模型,各组分别电针治疗7d后取材,观察各组血清乳酸脱氢酶(LDH)和超氧化物歧化酶(SOD)活性和心肌细胞氯离子通道CLC-3、CLC-4蛋白表达。结果:与模型组比较,内关组LDH水平降低(P<0.01)、SOD水平升高(P<0.01),心肌细胞氯离子通道CLC-3、CLC-4蛋白表达均明显减弱(P<0.01)。而列缺组、非经非穴组与模型组比较各指标均无明显差别。结论:循经电针内关穴对心肌缺血大鼠产生治疗作用,其机制之一可能是通过调节心肌细胞氯离子通道CLC-3、CLC-4蛋白表达而实现的。Objective: To study the effects of electro-acupuncture therapy along meridian on the expression of cardiac chloride channel CLC-3 protein and CLC-4 protein in myocardial ischemia rats induced by isoprenaline (ISO). Methods: The rat model of myocardial ischemia was induced by subcutaneous injection of the ISO. After electro-acupuncture therapy on the treatment groups for 7 days, the activity of lactate dehydrogenase (LDH) and superoxide dismutase (SOD) in serum and the protein expression of cardiac chloride channel CLC-3 and CLC-4 were detected in each group. Results: Compared with the model group, in Neiguan (PC6) group, the activity of LDH obviously decreased (P〈0.01), the activity of SOD obviously increased (P〈0.01) and the protein expression of cardiac chloride channel CLC-3 and CLC-4 reduced (P〈0.01). There was no significant difference among Lieque (L7) group, non-acupoint group and model group. Conclusion: The electro-acupuncture on Neiguan (PC6) along meridian had therapeutic effect on myocardial ischemia, mechanism of which may regulate the cardiac chloride channel CLC-3 protein and CLC-4 protein.
