摘  要:目的:观察对地鼠肾细胞生长及早期铅干预对新生儿神经行为的影响。方法:将生长状态良好的地鼠肾细胞用Versence-胰酶混合液消化后制成4×105浓度的细胞悬液接种24孔细胞培养板,每孔1mL,37℃二氧化碳培养。同时将苯酚按同样方法处理做为毒性对照组。受试产妇40例,受试组每日1剂,煎汤分3-5次服,10d为1个疗程。在3月龄时对研究对象用蓓蕾(Bayley)婴儿发育量表进行发育评价3个月±1周婴儿精神发育指数(MDI)和心理运动发育指数(PDI)。石墨炉原子吸收光谱法测定初乳铅含量,测定过程严格执行实验室内部质量控制。结果:经36℃连续观察5d未见细胞圆缩、肿胀、脱落等细胞毒性反应。40例婴儿3月龄时MDI、PDI两发育指标和杞金方药合剂早期铅干预间存在统计学意义。结论:用该方药合剂对地鼠肾细胞生长无不良影响,对新生儿神经发育有一定影响。Objective: To observe the effect of Qijin herbal on renal cells growth of hamster, and the influence of early lead intervention on neonatal neurobehavior. Methods: Hamster kidney cells were digested with pancreatic enzymes and then diluted to cell suspension with concentration of 4×105, and cultured in the cell culture plate, 1mL in each well, and cultured withcarbon dioxide at the temperature of 37℃. The control group of toxicity was dealing with phenol. 40 puerperas were subjected to experiment, and treated with Qijin herbal for 1 course(10 days). Bayley scales of infant development was used to evaluate the mental development index(MDI) and psychomotor developmental index(PDI) with 3 months old babies. Graphite furnace atomic absorption spectrometry was used to test the lead content in colostrums. Results: Observed for five days continuously, cells did not appear toxic reactions such as shrinkage, swelling and fall of. There was negative correlation between indicators of MDI and PDI and intervening measures of Qijin herbal and early lead intervention. Conclusion: Qijin herbal has no adverse effect on hamster kidney cells growth, and has some impacts on neonatal neurological development.
