艾灸血清对体外黑素细胞生长及SCF、c-kit蛋白表达的影响

摘　　要：目的:研究艾灸血清对体外培养人黑素细胞增殖、黑素合成,以及SCF、c-kit蛋白的影响,为艾灸治疗白癜风提供科学依据。方法:20只背部黄褐色毛豚鼠,随机分为艾灸组、对照组,豚鼠背部脱毛约2cm×2cm。艾灸组选用经外奇穴"灸癜风"和"阿是穴"作为艾灸治疗穴。穴上3cm处悬灸,采用温和灸补法,每日1次,每穴每次10min左右,疗程1个月;对照组动物同法固定,但不进行艾灸。末次治疗1h后,各组动物心脏采血,收集血清;采用体外培养正常人黑素细胞,观察艾灸血清、对照组血清在5%、10%、15%3种不同浓度,及24、48、72h,对黑素细胞增殖、黑素含量、酪氨酸酶活性,以及SCF、c-kit蛋白表达的影响。结果:艾灸血清可促进黑素细胞增殖、酪氨酸酶活性增加、黑素含量增加,并可促进SCF/c-kit信号通路中c-kit、SCF蛋白的表达。结论:艾灸血清中存在活性物质,可以促进黑素细胞增殖、酪氨酸酶活性增加、黑素含量增加,并可促进SCF/c-kit信号通路中c-kit、SCF蛋白的表达。Objective： To investigate the effect of moxibustion serum on the proliferation and synthesis of cultured human melanocytes in vitro, and the influence on SCF and c-kit protein expression, to provide a scientific basis for moxibustion treatment of vitiligo. Methods： 20 guinea pigs with brown fur in back were randomly divided into 2 groups. The hair was shaved approximately 2cmx2cm from the brown area on the dorsal skin of each animal. The moxibustion group was given the moxibustion on the points of ＇Jiudianfeng＇ and ＇Ashi＇ as treatment points, mild moxibustion should be away from the skin, about 3cm, and reinforcing method was used, once a day, 10min each time, the treatment course was one month. The animals in the control group were fixed by the same way, but without moxibustion treatment. One hour after the last treatment, each animal was taken blood from the heart, and the serum was collected. To observe the proliferation of melanocytes, melanin content, tyrosinase activity, and SCF, c-kit protein expression at the 24 hours, 48 hours, 72 hours and the serum concentrations of 5%, 10%, 15% by using normal human melanocytes in vitro. Results： Moxibustion serum could improve the melanocyte proliferation, tyrosinase activity and melanin content, which could promote the expression of SCF and c-kit protein. Conclusion： There are some active substances in the moxibustion serum, these substances can promote the melanocyte proliferation, tyrosinase activity and melanin content, which can promote the expression of SCF, c-kit protein in SCF/c-kit signaling pathway.
