中药清热利湿饮对HaCaT细胞增殖及凋亡的影响

摘　　要：目的:探讨中药清热利湿饮水提液对经肿瘤坏死因子(TNF-α)诱导活化的人永生化角质形成细胞(HaCaT)增殖及凋亡的影响。方法:采用MTT法检测清热利湿饮提取液对HaCaT细胞增殖的影响,采用流式细胞仪技术检测HaCaT细胞的凋亡。结果:25.00、50.00、100.00g/L清热利湿饮对经TNF-α诱导的HaCaT细胞均有明显的增殖抑制作用,与MTX阳性对照组相比,差异显著(P<0.05,P<0.01)。10.00g/L和12.50g/L清热利湿饮作用于活化后的HaCaT细胞,凋亡率为(44.94±4.16)%和(45.75±4.74)%,与MTX组(37.12±5.44)%相比有显著性差异(P<0.05)。结论:清热利湿饮可通过诱导细胞凋亡而抑制HaCaT细胞的过度增殖。Objective： To investigate the effects and effective concentration of Qingre Lishi Decoction extract on proliferation and apoptosis of keratinocyte HaCaT cells induced by TNF-α. Methods： The effects of Qingre Lishi Decoction on proliferation of HaCaT cells were observed by MTT method. The influence on apoptosis of HaCaT cells caused by Qingre Lishi Decoction was detected by l ow cytometry. Results： Concentration of 25.00, 50.00 and 100.00g/L of Qingre Lishi Decoctionhad a obvious inhibition on proliferation of HaCaT cells induced by TNF-α, and compared with MTX group, there was a significant difference（P〈0.05, P〈0.01）. The apoptosis of HaCaT cells treated by Qingre Lishi Decoction was（44.94±4.16）% and（45.75±4.74）%, and compared with MTX group（37.12±5.44）%, there was a significant difference（P〈0.05）. Conclusion： Qingre Lishi Decoction can inhibit the proliferation of HaCaT cells by inducing the apoptosis of cells.
