摘　　要：目的:通过对脾阴虚大鼠回肠水通道蛋白4(AQP4)表达分布的检测,探讨理脾阴正方对脾阴虚证大鼠水液代谢的影响。方法:对正常组、脾阴虚模型组、治疗组分别行免疫组织化学、RT-PCR及Western Blot的方法检测AQP4在回肠的表达及分布情况。结果:治疗组AQP4 mRNA及蛋白表达量均明显高于脾阴虚模型组(P<0.05);脾阴虚模型组AQP4 mRNA及蛋白表达量均明显低于正常组(P<0.05);脾阴虚模型组大鼠回肠上皮细胞细胞膜的AQP4分布量显著减少,治疗组其分布量有所回升,但仍低于正常组。结论:理脾阴正方可以在转录和翻译水平上调脾阴虚大鼠回肠AQP4表达,可能是其治疗脾阴虚证的作用机制之一。Objective： To discuss the effects of Lipi Yinzheng Formula （LPYZF） on water metabolism of rats with deficiency of spleen-yin syndrome by testing the distribution of aquaporin 4 （AQP4） of ileum of rats with spleen yin deficiency. Methods： Immunohistochemistry, RT-PCR and Western Blot were used to detect the expression and distribution of AQP4 on the ileum of rats. Results： The mRNA and protein expression of AQP4 in the LPYZR group were increased significantly than that in the model group （P〈0.05）. The mRNA and protein expression of AQP4 in the model group were decreased significantly than that in the control group （P〈0.05）. AQP4 expression in cell membrane was decreased significantly, however, after treatment by LPYZF, the distribution amount of AQP4 recovered, but it was still lower than that in the control group. Conclusion： LPYZF can increase the mRNA and protein expression of AQP4 in rats ileum with spleen yin deficiency at the level of transcription and translation. It may be one of the mechanisms in treating spleen yin deficiency syndrome.
