摘 要:目的:优化养正消积胶囊浸膏真空带式干燥的工艺参数。方法:采用HPLC-ELSD测定齐墩果酸含量,流动相乙腈-甲醇-0.5%乙酸(20∶70∶10)。以干膏含水率和齐墩果酸转移率为评价指标,采用正交试验考察进料速度、履带速度、加热系统温度对养正消积胶囊浸膏真空带式干燥工艺的影响。结果:最佳带式干燥工艺条件为履带速度6 cm·min-1,进料速度12 mL·min-1,干燥温度110℃,物料温度30℃,冷却区温度45℃,浸膏初始含固量约45%,真空度<-0.1 MPa;干膏含水率2.62%,RSD 1.2%;齐墩果酸转移率95.4%,RSD 0.42%。结论:优选的真空带式干燥工艺合理可行,干膏含水率低,质量好,适合养正消积胶囊的大生产推广。Objective:To optimize vacuum-belt drying process parameters of Yangzheng Xiaoji capsules extract.Method:HPLC-ELSD was employed to determine the content of oleanolic acid with mobile phase of acetonitrile-methanol-0.5% acetic acid (20∶ 70∶ 10).Taking water content of dry extract and transport rate of oleanolic acid as indicators,orthogonal design was adopted to investigate effects of feeding speed,belt speed and heating system temperature on vacuum-belt drying process of Yangzheng Xiaoji capsules.Result:Optimal parameters of vacuum-belt drying process were as follows:belt speed of 6 cm ·min-1,feeding speed of 12 mL· min-1 heating system temperature at 110 ℃ material temperature of 30 ℃ cooling zone temperature at 45 ℃ initial solid content in extract about 45%,vacuum degree <-0.1 MPa; moisture content of dry extract was 2.62% with RSD of 1.2% ; transfer rate of oleanolic acid was 95.4% with RSD of 0.42%.Conclusion:This optimized technology is reasonable and feasible with low water content and good quality,which is suitable for large scale production of Yangzheng Xiaoji capsules.
