摘 要:采用MTT法检测乌药根挥发油对7种人癌细胞系的细胞毒性以评价其体外抗肿瘤作用.利用乙醚萃取法、硅胶柱色谱法及GC(气相色谱法)和GC-MS(气质联用)法对乌药根挥发油的成分进行分离鉴定,并结合MTT法对分离得到的成分进行抗肿瘤活性筛选.结果表明:乌药根挥发油对所试肿瘤细胞均表现出明显的细胞毒性,特别是对人食道癌Eca-109细胞和人胃癌SGC-7901细胞增殖的抑制作用尤为明显(IC50=24.8μg/mL);经多种方法从乌药根挥发油中分离、鉴定并筛选到的抗肿瘤有效成分为吉马酮.The cytotoxic activities of Lindera strychnifolia root essential oil on seven human carcinoma cell lines were determined by using MTT assay. The components of root essential oil were separated by using ether extraction method, silica gel column chromatography, GC and GC-MS. And the antitumor active constituents were screened by using MTT assay. The results show that root essential oil exhibites significant cytotoxicity against all the carcinoma cells tested, and the human esophageal carcinoma (Eca-109) and human gastric carcinoma (SGC-7901) cell lines (IC50 : 24.8 μg/mL) are most sensitive to the cytotoxic effect of root oil. From several approaches, germacrone was obtained as cytotoxic constituent of Lindera strychnifolia root essential oil.
