摘 要:FBXL11属于连接酶SCF复合物中的底物蛋白识别组分F-box蛋白家族成员,为探明猪FBXL11基因的分子结构特征,研究采用生物信息学和分子生物学技术相结合的方法克隆鉴定猪FBXL11基因第18~ 20内含子,进行群体遗传、蛋白序列及进化特征分析.结果表明:含猪FBXL11基因第18 ~20内含子的克隆序列总长度为3 484 bp,第18、19和20内含子序列长度分别为561 bp、584 bp和442 bp,其编码蛋白质除具有典型保守F-box结构域之外,还有3个其他保守结构域.第18内含子的第377位存在SNP-TspEI (A/G),温氏集团长白猪种资源群体的遗传分析显示猪群具有杂合AG和纯合GG两种基因型,等位基因G在长白猪种中占有绝对的优势.同源基因FBXL11在物种间具有保守的分子特性,猪和牛属于进化系统树的同一组.猪FBXL11基因编码蛋白包含4个保守结构基序,第18内含子在长白猪种资源群体中存在遗传差异,FBXL11蛋白具有维持基因组稳定及调控细胞分化的潜能.FBXL11 belongs to a member of F-box protein family in the recognizing components of substrate protein in ligase SCF complex. In order to explore the molecular structural feature of porcine FBXLll gene, the 18th -20th intron of porcine FBXLll gene was cloned and i- dentified, moreover, group genetic analysis, protein sequence and evolution character analysis were conducted by combining bioinformatics and molecular biological techniques in this study. The results showed that the total cloning sequence with 18th ~ 20th intron of porcine FBXL11 gene was 3 484 bp long, and the sequence length of the 18th, 19th and 20th intron was 561 bp, 584 bp and 442 bp, respectively. In addition, its coding protein comprised three other conservative structural domains, besides a typical conservative F-box. SNP-TspEI (A/ G) was identified at the 37Th site in the 18th intron of porcine FBXLll gene. The genetic analysis on TspEI-RFLP in pure Landrace breed resources from Wen's Group indicated that there were two different genotypes including heterozygous AG and homozygous GG, and allele G was absolutely in dominance. Homologus FBXL11 gene shared the conserved molecular trait among interspecies, only Sus Scrofa and Bos Taurus belonged to the same group from the phylogenetic tree. Porcine FBXL11 gene coded a protein containing four conservative structural domains, and the genetic diversity of the l8th intron appeared in Landrace breed resources. FBXL11 protein could maintain the stabilization of genome and had potentiality in regulating cell differentiation.
