摘  要:目的探讨过氧化物酶增殖物激活受体-γ(PPARγ)和核因子E2相关因子-2(Nrf2)在肾脏衰老中的作用。方法以3个月龄和24个月龄大鼠作为青年组和衰老组(n=10),PAS染色观察肾脏组织病理改变,肾脏组织匀浆测定H2O2、丙二醛(MDA)含量,Western Blot法检测肾脏组织髓过氧化物酶(MPO)、PPARγ和Nrf2蛋白质表达。结果衰老组大鼠肾脏组织MDA[(44.42±4.17)μmol/mg比(26.23±1.43)μmol/mg]、H2O2[(239.90±39.38)μmol/μg比(110.00±19.49)μmol/μg]含量较青年组均明显增加,差异有统计学意义(P<0.01、P<0.05);衰老组肾脏组织MPO蛋白质表达较对照明显升高,差异有高度统计学意义(P<0.01),而PPARγ蛋白表达则明显下降,差异有高度统计学意义(P<0.01)。两组间Nrf2蛋白表达差异无统计学意义(P>0.05)。结论氧化损伤在肾脏衰老中发挥重要作用,可能是通过PPARγ表达下降介导。Objective To observe the role of peroxisome proliferator-activated receptor gamma(PPARγ) and nuclear factor- E2- related factor-2(Nrf2) on the kidney aging. Methods Rat aging model was established using 3- month and24-month old rats as young group and aging group(n = 10). Renal pathological changes were tested by PAS reaction.The amount of the H2O2 and MDA were tested using kidney homogenate. Western blotting was used to determine the protein expression of MPO, PPARγ and Nrf2. Results Renal MDA [(44.42±4.17) μmol/mg vs(26.23±1.43) μmol/mg]and H2O2[(239.90±39.38) μmol/μg vs(110.00±19.49) μmol/μg] of aging group were significantly higher than those of young group(P 〈0.01 and P〈 0.05). Western blotting assay showed that the renal expression of MPO protein of aging group increased apparently compared with young group(P〈0.01), but PPARγ decreased significantly compared with young group(P〈 0.01). And the expression of renal Nrf2 had no significant difference between two groups(P〉 0.05).Conclusion Oxidative damage plays a role on the kidney aging, which is correlated with the decreasing of PPARγ expression of renal tissue.
