稳斑汤对ApoE基因敲除小鼠主动脉粥样硬化不稳定斑块HSP70表达的影响

摘　　要：目的观察搜风祛痰法中药复方稳斑汤对ApoE基因敲除小鼠主动脉粥样硬化(AS)不稳定斑块HSP70表达的影响。方法 ApoE基因敲除小鼠高质饮食喂养13周后,待其形成成熟的AS斑块,随机分为模型组、稳斑汤低剂量组、稳斑汤中剂量组、稳斑汤高剂量组、西药组、空白对照组。各组相应处理13周后,取主动脉组织HE染色后在光学显微镜下进行病理学观察,然后采用免疫组化和RT-PCR技术检测HSP70的表达水平。结果治疗组主动脉组织中HSP70表达水平高于对照组(P<0.05或P<0.01),稳斑汤高剂量组HSP70表达水平与西药组无明显差异(P>0.05)。结论捜风祛痰中药复方稳斑汤可通过影响HSP70以稳定易损斑块,其机制可能与HSP70对心血管系统的保护作用相关。Objective： To explore the effect of compound stablizing plaque decoction on unstable plaque HSP70 （Heat Shock Protein 70） expression of ApoE knockout rats with AS. Methods： 70 ApoE knockout mice were fed a high-fat diet for 13 weeks until the mature atherosclerotic plaques were formed. Thereafter,they were randomly divided into the model group,low-dose, medium-dose, high-dose of stabilizing plaque decoction groups,western medicine control group,and normal group. After 13 weeks,aortic tissues of each group were fetched to observe under an optical microscope after HE staining. And then immunohistochemical methods and RT-PCR technique were used to detect the level of HSP70 expression. Results： The HSP70 expression levels in aortic tissue of mice in the treatment groups were higher than control group （P〈 0.05 或 P〈 0.01 ）,and the expression levels of HSP70 between TCM high-dose group and western medicine group had no significant differences（P〉 0.05 ）. Conclusion： Searching wind and eliminating phlegm compound stabilizing plaque decoction may stabilize plaques by influenc- ing the HSP70, and its mechanism may be associated with its protection for the cardiovascular system.
