摘  要:目的通过观察粒细胞集落刺激因子(G-CSF)对脑出血(ICH)大鼠血肿周围血管内皮生长因子(VEGF)表达的影响,探讨脑出血后G-CSF发挥神经保护作用的途径。方法 105只SD大鼠随机分为假手术组、ICH组和治疗组,每组各35只。断尾取自体血法制备ICH模型,造模1 h后治疗组经腹腔注射重组G-CSF(60μg/kg),假手术组、ICH组经腹腔注射等量等渗盐水。各组大鼠术后6 h、24 h、48 h、72 h、7 d、14 d、21 d进行神经功能障碍评分,处死大鼠,运用免疫组织化学方法检测血肿周围VEGF表达。结果治疗组大鼠从24 h开始各时间点神经功能障碍评分较ICH组明显增加(P<0.05)。假手术组仅见少许VEGF表达;ICH组VEGF表达从6 h开始出现增多,7~14 d达到高峰;治疗组各时间点VEGF表达较ICH组明显增多(P<0.05),72h^7d到达高峰。结论 G-CSF可能通过上调脑出血血肿周围VEGF表达发挥神经保护作用。Objective To investigate the effect of granulocyte colony-stimulating factor on expression of vascular endothelial growth factor in rat brain with intracerebral hemorrhage, and to explore its possible neuroprotective effects. Methods One hundred and five Spragne Dawley rats were randomly selected and divided into 3 groups: sham operation (35), ICH group (35) and treatment group (35). The rat experimental ICH model was built up by infusing fresh autologous blood. Rats in the treatment group were administered rhG-CSF (60 μg/kg) by intraperitoneal injection post operation lh, while rats in ICH group and sham operation group were injected with equal volume of isotonic saline solution. Neural function defect of rats were assessed by measurement of Garcia's score method post operation 6h, 24h, 48h, 72h, 7d, 14d, 21d.Then the rats were sacrificed, expressions of VEGF around hematoma in rats' brain were analyzed by immunohistochemistry method at the same time. Results The score of nerve function was obviously increased from 24h in treatment group, and had significant difference compared with ICH group (P〈0.05). The expression of VEGF was little in sham operation group. In ICH group, the expression of VEGF increased from 6h and peaked at 7d-14d. Compared with the ICH group, the expression of VEGF in treatment group increased at each time points (P〈 0.05) and peaked at 72h-7d. Conclusion G-CSF could up-regulate expression of VEGF around the hematoma to exert neuro-protective effects.
