内脂素与NF-κB在子痫前期发病中的作用研究

摘　　要：目的测定内脂素、核因子κB(NF-κB)在子痫前期孕妇胎盘、血清及其胎儿脐血中的表达情况,探讨子痫前期发病与炎症的关系。方法选择子痫前期患者40例作为患者组,正常同孕周孕妇40例作为对照组。采用酶联免疫吸附双抗体夹心(ELISA)法检测2组孕妇血清及胎儿脐血中内脂素和NF-κB的水平,采用免疫组化(S-P)法检测胎盘中内脂素和NF-κB水平。结果患者组孕妇胎盘、血清中内脂素及NF-κB含量均较对照组明显升高(P均<0.05);患者组孕妇血清及胎盘中内脂素和NF-κB之间存在正相关性(r=0.67,P<0.01)。2组胎儿脐血中内脂素及NF-κB含量比较差异无统计学意义。结论内脂素可能作为一种前炎性因子通过增加炎症应答转录因子NF-κB的转录能力来参与多种血管炎症过程,引起血管内皮细胞损伤,导致子痫前期的发病。Objective It is to determine the expression of visfatin and nuclear factor κB（ NF- κB） in fetal umbilical cord blood,blood serum and placenta in preeclampsia pregnant women as well as to discuss the relationship between inflammation with the onset of preeclampsia. Methods 40 cases of preeclampsia patients were selected as patient group and 40 cases of normal pregnant women as the control group. The contents of visfatin and NF- κB in blood serum and fetal umbilical cord blood were determined by enzyme-linked immunosorbent double antibody sandwich（ ELISA） method and the contents in placenta by immunohistochemical method in the pregnant women in both groups. Results The contents of visfatin and NF- κB in blood serum and placenta in the pregnant women in patient group were obviously higher than that in the women in control group（ P 〈0. 05）,there were positive correlations between visfatin and NF- κB in blood serum and placenta in the women in patient group（ r = 0. 67,P 〈0. 05）. There was no significant difference in the contents of visfatin and NF- κB in fetal umbilical cord blood in both groups. Conclusion Visfatin may be as a former inflammatory factor by increasing the inflammatory response transcription factor NF- κB transcription ability to participate in a variety of vascular inflammation process,thus to cause endothelial cell damage and lead to the onset of preeclampsia.
