摘 要:目的探讨通络救脑注射液保护神经元的可能作用机制。方法制备小胶质细胞氧糖剥夺条件液、通络救脑条件液、栀子苷条件液、人参皂苷Rg1条件液,将神经元分为正常对照组、氧糖剥夺组、正常条件液组、氧糖剥夺条件液组、通络救脑条件液组、栀子苷条件液组、人参皂苷Rg1条件液组,其中正常对照组采用常规神经元培养液,氧糖剥夺组采用无糖DMEM培养液,其余5组按照处理条件分别使用对应的小胶质细胞条件培养液,条件培养液添加为100%,作用时长为6h。比较各组神经元细胞活性、乳酸脱氢酶(LDH)漏出率、神经元NMDAR1及caspase-3的表达水平。结果与正常对照组比较,氧糖剥夺组和氧糖剥夺条件液组神经元细胞活性明显下降,LDH漏出率明显增加,神经元NMDAR1和caspase-3表达均明显上升(P<0.05或P<0.01)。与氧糖剥夺组比较,通络救脑条件液组、栀子苷条件液组、人参皂苷Rg1条件液组神经元细胞活性明显提高,通络救脑条件液组和栀子苷条件液组LDH漏出率降低、NMDAR1表达明显下降(P<0.05或P<0.01),通络救脑条件液组、人参皂苷Rg1条件液组caspase-3表达明显下降(P<0.05或P<0.01)。结论通络救脑注射液及其单体成分(栀子、三七有效组分)可通过调节小胶质细胞的分泌成分,降低LDH漏出率及神经元NMDAR1、caspase-3的表达水平,提高神经元活性,逆转损伤小胶质细胞可能对神经元造成的影响,从而发挥神经元的保护作用,并且通络救脑注射液有整合调节的治疗优势。Objective To investigate the possible action mechanism of Tongluo Jiunao Injection in protecting neurons. Methods The microglia glucose deprivation (OGD) conditioned medium, Tongluo Jiunao conditioned medium, geniposide conditioned medium and ginsenoside-Rgl conditioned medium were prepared. The neurons were classified into the normal control group, OGD group, normal conditioned medium group, OGD conditioned medium group, Tongluo Jiunao conditioned medium group, geniposide conditioned medium group and ginsenoside-Rgl conditioned medium group. In normal control group, neurons were cultured in conventional way, OGD group was treated with glucose-free DMEM culture and the rest 5 groups were cultured in the responding microglia conditioned mediums.The conditioned mediums were added as 100% and the action time was 6 hours. The cell viability, leakage rate of lactate dehydrogenase Results Comparing rate was significantly OGD group and OGD (LDH) and expressions of NMDAR1 and caspase-3 of neurons in each group were compared. with the normal control group, the neuron viability was significantly decreased, LDH leakage increased and expressions of neuron NMDAR1 and caspase-3 were significantly increased in conditioned medium group (P 〈 O. 05 or P 〈 O. 01 ). Comparing with the OGD group, the neuron viability in Tongluo Jiunao conditioned medium group, geniposide conditioned medium group and ginsenoside-Rgl conditioned medium group was significantly increased, LDH leakage rate and expression of neuron NMDAR1 was sig- nificantly decreased in Tongluo Jiunao conditioned medium group and geniposide conditioned medium group (P 〈 0. 05 or P 〈0. 01 ) and the expression of caspase-3 was significantly decreased in Tongluo Jiunao conditioned medium group and ginsenoside-Rgl conditioned medium group ( P 〈 0. 05 or P 〈 0. 01 ). Conclusion Tongluo Jiunao Injection and its monomer composition (efficient compositions of gardenia and panax notoginseng) can decrease LDH leakage rate and expressions of NMDA
