摘  要:目的研究过氧化甲乙酮(MEKP)对大鼠肝脏的氧化损伤作用。方法 50只SD雄性大鼠随机分成3个MEKP染毒组(50、500、1 000 mg/m3)和1个溶剂对照组(2,2,4-三甲基-1,3-戊二醇二异丁酸脂)、1个空白对照组,每组10只,每周吸入染毒5d,每天6 h,13周后观察肝脏的组织形态学损伤,测定肝脏组织的还原型谷胱甘肽(GSH)、丙二醛(MDA)含量和超氧化物歧化酶(SOD)活力。结果 500、1 000 mg/m3剂量染毒组大鼠肝小叶周围汇管区出现细胞核浓缩、脂肪变性,肝细胞轻度水肿及淋巴细胞浸润等病理学变化。500和1 000 mg/m3组肝脏组织MDA水平升高,SOD活力下降,GSH含量下降,差异均有统计学意义(P<0.05)。结论 MEKP可诱导大鼠肝脏组织氧化应激损伤及组织病理学结构的改变。Objective To investigate the oxidative damages of methyl ethyl ketone peroxide (MEKP) on liver tissue of male rats. Methods A total of 50 healthy male SD rats were randomly divided into five groups (10 in each group), which were exposed to 50, 500 and 1 000 mg/m^3 aerosol of MEKP, and 2, 2, 4-trimethyl-1, 3-pentanediol isobutyrate (solvent control group), and one bland control group respectively, via whole body inhalation for 6 h per day, 5 d per week,13 weeks, then the histological change of the liver was examined and the contents of glutathione (GSH) , malondialdehyde (MDA), the activities of antioxidants enzymes superoxide dismutase (SOD) in rat liver were assayed, respectively. Results The histopathological changes of rat livers were observed in 500 and 1000 mg/m^3 groups, such as nucleus condensation and fatty degeneration in the periportal zone of the hepatic lobule, mild edema of the liver cells, and lymphocyte infiltration. The MDA level was significantly higher and the level of GSH, the activity of SOD were significantly decreased with the increase of exposure doses, the difference between 500, 1000 mg/m^3 group and control group was statistically significant (P〈0.05). Conclusion MEKP can cause oxidative damage and histopathological structure change in the liver of rats.
