调气行水合剂联合腹腔注射顺铂IL-2治疗肝癌腹水的实验研究

摘　　要：目的:研究调气行水合剂结合顺铂、IL-2腹腔注射对肝癌腹水小鼠模型治疗作用及细胞凋亡基因Bax、Caspase-3蛋白表达的影响,从分子生物学水平揭示其治疗肝癌腹水的机制及其效果。方法:将72只纯系昆明小鼠随机分为对照组,纯中药组,中西医结合大、中、小剂量组及西药组。用药结束后,处死小鼠,测量腹水量、瘤重,记小鼠生存期。应用免疫组化法检测小鼠肝肿瘤组织Bax、Caspase-3蛋白表达情况。结果:中西医结合组腹水量明显减少,较对照组及西药组比较差异显著。中西医结合组小鼠瘤重明显小于对照组,差异有显著性。生存期:中西医结合组、中药组、西药组均较对照组明显延长,差异有统计学意义。中西医结合大剂量组细胞凋亡基因Bax蛋白的阳性表达率显著高于对照组(P<0.01)。中西医结合大、中剂量组细胞凋亡基因Caspase-3蛋白的阳性表达率显著高于对照组(P<0.01)。结论:中西医结合治疗可减少腹水量,减轻瘤重,延长生存期。中西医结合大剂量组可明显增加细胞凋亡基因Bax蛋白表达,中西医结合大、中剂量组可明显增加细胞凋亡基因Caspase-3蛋白表达,诱导细胞凋亡。揭示了调气行水合剂联合顺铂、IL-2腹腔注射治疗肝癌腹水的作用机制。Objective ： To investigate the effect of Tiaoqi Xingshui Mixture combined with peritoneal injection DDP and IL-2 on mouse with ascetes hepatoma, to explore the mechanism of the treatment for Ascetes hepatoma on the level of molecular biolo- gy. Methods： 72 pure hnes of Kunming mice were randomly divided into controlled group, Chinese medicine group, combina- tion of traditional Chinese and Western medicine, in high, middle, low dose group and western medicine group. After the medi- cation, the mice were killed. Measure the volume of ascetes tumor burden and life-span. Immunohistochemical method was used to detect the mouse liver tumor tissue Bax, Caspase-3 expression protein. Results： The volume of ascetes were markedly reduced in high dose group of TCM combined with western medicine. The difference were significant compare with controlled group and western medicine group. Tumor burden markedly decrease in the group of TCM combined with western medicine than the controlled group. The life-span prolong obvious in group of TCM combined with western medicine, Chinese medicine group and western medicine group. The difference were significant compared with the controlled group. The positive expression of apoptosis gene Bax protein in high dose group of TCM combine with western medicine was higher than that of the controlled group （ P 〈 0. 01 ）, and the positive expression of apoptosis gene Caspase-3 protein in high and middle dose group of TCM combined with western medicine was higher than that of the controlled group （P 〈0. O1 ）. Conclusion： Treating ascetes hepatoma with TCM and western medicine can reduce the volume of ascetes, decrease tumor burden and prolong life-span. The high dose group of TCM combined with western medicine can significantly increase the expression of apoptosis gene Bax protein. The high and middle dose group of TCM combined with western medicine can significantly increase the expression of apoptosis gene Caspase-3, protein, induction of apoptosis. Bring the light to the me
