盐酸氨溴索对肺间质纤维化大鼠支气管肺泡灌洗液头孢噻肟浓度的影响

摘　　要：目的探讨盐酸氨溴索对肺间质纤维化大鼠支气管肺泡灌洗液(BALF)头孢噻肟浓度的影响。方法将54只Wister雄性大鼠随机分成3组,每组18只,正常对照组(A组)、模型组(B组)、盐酸氨溴索组(C组),B、C组采用气管内注入博来霉素(5 mg/kg)的方法制作肺间质纤维化动物模型,A组气管内灌注生理盐水。C组每天腹腔内注入盐酸氨溴索35 mg/kg。分别于给药后第7、14、28天,各组每次取6只大鼠,鼠尾静脉注射头孢噻肟(600 mg/kg)后行支气管肺泡灌洗,灌洗液采用液相色谱—质谱联用法测定头孢噻肟的浓度,并取肺组织做病理观察。结果第7天C组BALF头孢噻肟浓度低于B组,差异无统计学意义(P=0.164),第14天C组BALF头孢噻肟浓度上升,C组与B组的差异有统计学意义(P=0.000)。第28天C组锐降,低于B组,差异无统计学意义(P=0.126)。结论盐酸氨溴索可提高肺纤维化初期BALF中头孢噻肟的浓度。Objective To explore the effect of ambroxol on the concentration of cefotaxim in bronchioalveolar lavage fluid（BALF） on rats with bleomycin-induced pulmonary fibrosis in rats.Methods A total of 54 Wister male rats were randomly divided into three groups,including normal control （Group A）,model（Group B）,ambroxol-treated （Group C）.On experimengtal day （day 0）,the rats was intratracheally instilled with bleomycin（5mg/kg body weght） or sterile saline,and then treated with the ambroxol（35 mg/kg） or sterile saline intraperitoneally respectively.On day 7,14,28 after instillation,six rats of each group were sacrificed and the bronchial alveolar fluids were recovered and the lung were gotted for histopathological examination after injected cefotaxim（600 mg/kg）intravenously.The concentration of cefotaxim in the BALF was assayed by liquid chromatography-mass spectrometry method.Results On 7th day the concentration of cefotaxim in the BALF of ambroxol group was lower than model group.On 14th day the concentration of cefotaxim in the BALF of ambroxol group was higher than model group.The deference between ambroxol group and model group was significant statistically （P =0.000）.On 28th day the concentration of cefotaxim in the BALF of Ambroxol group decreased sharply,and was lower than model group （P =0.126）.Conclusions Ambroxol can increase the concentration of cefotaxim in the BALF of the primary fibrosis stage.
