摘 要:目的探讨siRNA干扰SUMO-1基因表达对人子宫内膜癌Ishikawa细胞增殖的影响,旨在为子宫内膜癌基因治疗提供新的靶点。方法设计针对SUMO-1基因的siRNA转染Ishikawa细胞;采用实时荧光定量RT-PCR和Western-blot检测转染效果;MTT及流式细胞术检测转染siRNA后,细胞的增殖及周期情况。结果转染SUMO-1 siRNA明显抑制Ishikawa细胞增殖,细胞周期被阻滞于G1期,由G1期向S期进展延缓(P<0.05)。结论 SUMO-1 siRNA能有效抑制Ishikawa细胞增殖,SUMO-1有望成为子宫内膜癌基因治疗新途径。【Objective】To investigate the effect of SUMO-1 gene on proliferation in human endometrial cancer Ishikawa cell in vitro, and to provide a new target for endometrial cancer gene therapy. 【Methods】SUMO-1 gene siRNA transfected Ishikawa cells and the transfection effects were detected by Real-time quantitative RT-PCR and Western-blot. The proliferation and cycle of the cells were evaluated by the MTT and the Flow cytometry.【Results】After transfected with SUMO-1 siRNA for 48 hours, Ishikawa cell proliferation was inhibited, the G1 phase was blocked and the cells phase from G1 to S phase was delayed(P〈0.05). 【Conclusions】SUMO-1 siRNA reduced Ishikawa cells proliferation. It might be a new gene therapeutic target for human endometrial cancer.
