摘 要:为了探索miRNA在文冠果不同类型植株的花发育阶段不同组织的表达特异性,以单瓣型和重瓣型文冠果的花芽、茎、叶为试材,提取这2种类型组织的小RNA。从前期研究获得的文冠果小RNA测序数据库中筛选11个候选miRNA,采用实时荧光定量PCR法检测其在花发育不同阶段以及茎和叶片的表达特点。结果表明,11个候选miRNA具有不同的表达形式。Xso-M007a-b、miR167a-b与miR169在两类中都是表达量随花芽发育呈现下降趋势,XsoM003则相反,说明了miRNA表达的时序性。在不同类型之间以及不同部位miRNA的表达也存在差异,Xso-M002、Xso-M003、Xso-M007a-b、miR169、miR319和miR827在两类型花芽中的表达量均高于茎和叶中的表达量。对不同类型、部位和时间点上的表达情况分析,表明miRNA具有明显的品种特异性、组织特异性和时序性。此外,结合miRNA的功能与其表达量差异分析发现,文冠果的形态结构特征与miRNA的表达量变化密切相关。为探索miRNA表达量的变化在林木花器官形成与不同组织分化中的作用提供了重要信息。The aim of this paper is to explore miRNA expression specificities in different flower development stages and tissues in different types of Xanthoceras sorbifolia. Flower buds,stems,and leaves of single and double flower types were taken as materials. High quality small RNAs were isolated from four tissues of the two types. 11 candidate miRNAs were chosen from small RNAs data bank of Xanthoceras sorbifolia,and qRT-PCR was used to analyze the miRNAs expression in different flower development stages,stems and leaves. The result showed 11 miRNAs had different expression characters. The expression trends of Xso-M007a-b,miR167a-b and miR169 were down-regulated with the flower buds development in two types; on the contrary Xso-M003 was up-regulated. This demonstrated the sequential expression of miRNA. There was also differential expression among different types and tissues. In both types Xso-M002,Xso-M003,Xso-M007a-b,miR169,miR319 and miR827 showed higher expression in flower buds than in stems and leaves. Expression analysis on different types,tissues and time points proved the miRNA's expression specificities: species-specificity,tissue-specificity and time-specificity. In addition,based on miRNA function and expression difference,it was found that morphological characteristics were closely related to miRNA expression. The paper provided valuable information for exploring the function of miRNA expression variation in forest flower formation and tissues differentiation.
