摘 要:为保护苦苣菜野生资源,以苦苣菜下胚轴为材料,进行了愈伤组织诱导培养、分化培养、生根培养和试管苗移栽及定植的研究,结果表明:MS+琼脂4.8 g·L-1+蔗糖42 g·L-1+KT 0.4 mg·L-1+NAA 0.2 mg·L-1+2,4-D2.0 mg·L-1是愈伤组织诱导生长的适宜培养基;1/2 MS+琼脂4.5 g·L-1+蔗糖35 g·L-1+ZT 0.8 mg·L-1+NAA 0.1 mg·L-1是愈伤组织块分化培养的适宜培养基;1/4 MS+IAA 0.6 mg·L-1+琼脂4.3 g·L-1+蔗糖15g·L-1是生长芽试管苗培养的适宜培养基;温室内试管苗移栽成活率为91.7%;试管苗在山林旁定植成活率为97.1%,定植试管苗的植物学性状与实生苗基本一致.In order to preserve the wild resources, with the method of tissue culture, the hypocotyls of Sonchus oleraceus L. were used as materials to do the research on the cultivation of callus inducement, differentiation and rooting, transplantation of tube seedlings and stable planting. The results demonstrated that MS+Agar 4.8 g·L^-1+Sucrose 42 g·L^-1 + KT 0.4 mg·L^-1 + NAA 0.2 mg·L^-1 + 2,4-D2.0 mg·L^-1 was the ideal medium for callus inducement; 1/2 MS + Agar 4.5 g·L^-1+Sucrose 35 g·L^-1+ ZT 0.8 mg·L^-1+ NAA 0.1 mg·L^-1 was the optimum medium for callus differentiation cultivation;The ideal medium for tube seedlings cultivation was 1/4 MS+IAA 0.6 mg·L^-1+ Agar 4.3 g·L^-1+ Sucrose 15 g·L^-1. The transplanting survival rate of tube seedlings was 91.7% in greenhouse. The planting survival rate of tube seedlings was 97.1%. The tube seedlings stable planted maintained all botany characteristics of the wild Sonchus oleraceus L.
