摘 要:目的:建立IGT的病证结合大鼠模型,为后续研究奠定基础。以高脂饲料长程喂养结合限制活动空间方法,模拟人类"多食少动"不良生活方式,复制大鼠IGT模型,从表观特征和相应生化指标探讨该模型的"痰湿瘀热"证候属性。方法:采用长程(20w)以45%脂肪热量比标准高脂饲料(对照饲料为10%脂肪热量比标准饲料)结合限制活动空间的喂养方式,复制大鼠IGT模型,通过体质量、腹围、体长、活动度、毛色、二便情况、血液颜色及黏稠度等表现结合血糖、血清胰岛素、血脂、血液流变学等指标,探讨IGT模型的"痰湿瘀热"证候属性。结果:造模结束时,高脂饲料组较之对照饲料组,大鼠嗜卧少动、腹型肥胖明显,毛色暗淡稀疏,小便黄、大便溏且异味明显,血液颜色瘀黯而黏稠。与对照饲料组比较,高脂饲料组空腹血糖无明显变化,而餐后2h血糖增高明显,与对照饲料组比较差异显著(P<0.01)符合IGT诊断标准;高脂饲料组大鼠Lee's指数增加,血脂增高,全血粘度、血浆粘度增加而红细胞变形能力下降,以上指标与对照饲料组比较差异显著(P<0.01)。结论:此种模型复制方法得到的IGT大鼠模型,从表观特征以及实验室指标方面基本符合中医"痰湿瘀热"特征,该模型为后续中医药防治IGT的研究奠定模型基础。Objective:To investigate a kind of IGT rat models which combined disease and syndrome together, it could help lay the foundation for further study.We used the method of long-term high-fat diet and activity space limitation to simulate the physical inactivity and overeating unhealthy human lifestyle,in order to copy the IGT rat models.We dis-cussed the properties of the models with phlegm-dampness stagnate heat symptom from both phenomenal characteristics and related biochemical indexes.Methods:We used the twenty-weeks-long high fat diet feeding and activity space limitation method to replicate the IGT rat models.Lipid energy percentage of high fat diet and standard diet were 45%and 10 %.We analyzed the properties of the models with phlegm-dampness stagnate heat symptom from two sides.One side was the expressions of weight ,abdominal circumference ,body length ,activities ,hair color, urine and stool ,blood color and viscosity.The other side was the indexes of blood glucose, serum insulin, blood fat and hemorheology.Re-sults:After modeling, the rats in high fat diet group addicted lying and became in abdominal obesity shape obviously;their hair colors became pale and sparse;they also expressed yellow urine, loose stool with bad smell and blood became dim color and sticky when compared with the standard group.The fasting blood-glucose of rats in high fat diet group showed no significant difference, but two hour post -meal blood glucose increased clearly when compared with the standard group(P〈0.01).These expressions fitted the diagnostic criteria of IGT .Lee's indexes, blood fat,the whole blood viscosity, plasma viscosity of high fat diet group increased ,but red cells deformation ability decreased,and these indexes showed great significances when compared with the standard group( P〈0 .01 ) .Conclusion:The IGT rat models which are replicated by this method are largely in line with the phenomenal characteristics and related biochemical inde-xes of the phlegm-dampness stagnate heat symptom in TC
